Sterilization Question

Hey Everyone! Newbie here. Second attempt, different set of syringes. First attempt - 24 jars, zero signs of (the proper looking per MMGG) mycellium. I am about to start sterilizing the jars for the second try and I have 2 ?s:
1. Do I need to STERILIZE (PC) the jars prior to sterilizing them with the substrate inside?
2. In the PC, should I include about an inch of H2O in the bottom during sterilization?

You made no mention as if ANYTHING grew in your jars. If no containaments were present, you must be sterlizing OK. You want enough water in the pc to cook for about 45 minutes to an hour (everyone says at 15 psi/ word has if that 10 psi works just as well). You want the water a little below the rim of the jars (about 3/4' to an inch below the rims of the lids). If after inoculation nothing grows in the jars after a couple of weeks there are two obvious posibilitys. First you may be produceing too dry a substrait by useing too little water in the jars/lids on too lose in the pc thus loseing water content to envaporation. Secondly, this is fairly common: If you flame the needed of the syringe between jars, as so many do here, you have to be very carefull to let the needle cool down enough before useing it.... it might take over two or three minutes... five minutes and you know it's cooled enough to be swabed with alcohol just before inoculation. You can EASILY kill all of the spores in a syringe by inoculating with a hot needle! Good luck.

Okay, thanks guys. In the first try there was lots of off-color mycellium on some, nothing on the others. Additionally, it took about 3-4 weeks to see anything! 70-85 degrees 65% humidity.
So I just sterilize in a dry PC after I've added the substrate to washed and dried jars?

would it really kill all of the spores or just what comes out at the time. that what i think cuz that happened to AFOAF. he inncolualted 10 jars with one syringe. some of the jars colonized in all four holes but he remembers that the last few he had to do fast and probably did not let the syringe cool enough so a few just colnized in one or two spots not four. hope i helped

Here is an example of what I meant: If one was to flame the syringe everytime between jars and not wait more than a few seconds, all of the spores could be killed. It's very common. Some dont even bother with flameing before inoculation, but I dont recomend it.

I guess i'd be one of those people.. :?
I just use an alcohol soaked cotton swab each time.. seems to work fine for me

NO, infadel. Never sterilize in a dry PC! Read Jammer's instructions again. There should be water in the bottom of the PC. The bottom jars should be sitting in water with water about half way up the sides. Heating a dry PC would cause the metal to over heat and blow the safty valve and destroy your jars.

i think the first reply thought you were asking if you need to but H202 in the PC (thats what i saw it as first).
always have water in a PC
