Well, this has been a busy week for me. As some may know from my other post I just successfully completed my first G2G with great results. Now, my LC's have also tested good.
I started with 12 1/2 pints of the light Karo TEK. Started out a little wrong right off the bat. Used 1 tablespoon Karo per 1/2 pint which only contained about 6 ounces water so my LC was a little sweet to say the least. Pc'ed at 15 psi for 30 minutes. Let sit overnight and multi spore inoculated on 4-16.
Out of the 12, 2 grew something black and funky the next day. Knew this was way to soon for any kind of shroom growth from multi spore so was pretty sure those two were trash. Didn't look like it was clumping spores either. Stashed these two away just to see what would grow. Kinda like looking at a car wreck. Probably not going to be pretty, but you can't keep from looking. Well, pulled them out today and this is what they look like,
Don't look near as bad as they did a month ago. Might shoot up a jar with them just to check them out. Never know.
Besides those two, had 5 that did nothing. No contams or growth of any kind. Maybe too sweet? I'll open those in the glovebox and throw some colonized grain in them and see what happens.
The surviving 5 seem to have come out just fine. Inoculated 5 jars with them on 5-21, more like early morning 5-22 and this is what they look like 5 and a half days later.
Wasn't too impressed after the first two days. There was growth visible, but, you know, not a whole lot. They really took off the last couple of days. Almost fully colonized in under 6 days. Yes, I'm impressed. Between this and G2G, I'm wondering why I needed to learn patience,lol. I guess this all seems really fast after doing cakes. Another good reason to start with cakes. Those bitches will teach you all about patience. Well, be careful with that sugar, honey, Karo or whatever. Too much isn't a good thing. I'm using malt extract and dextrose now. Have a few of those going now. let you all know how it comes out later. Well, guess I have to recommend this right up there with G2G. Isolates next. Gotta keep on keeping on. Peace.

I just knocked up my agar tonight/morning. You have good results from LC, my LC is so so sososososososos slow, i dunno why. its just slow.

thanx for the post hippiechick, i'm also experimenting with LC. how much time and at what temperature did those jars incubate before looking like that? my honey bottle is on his day 10 right now (room temp) and only has some small white things floating in it.

Mine were slow also. These were done on 4-16, almost a month and a half ago, and are now just ready to go in the fridge. They were incubated at 82 degrees.

Yea you probably had too much Karo in the ones that didn't work, I've had that happen. Guess its like killing fish if you give them too much food.

I believe so. You know though, they have lightened up in color by about 50%. I'm still gonna toss some colonized grain in them to see what happens. 5 worked with multispore so it is possible for them to sustain growth. Maybe already colonized mycelium will have a better chance then spores. After all, mycelium can handle H2O2 , but spores can't. Let you know what happens. Peace,Love,Happiness and Harmony. I have just took a go at makeing my first LC using the baby bottle tek with only one problem when I pulled the needle out of one of the nipple the silicone that I used to make the self healing injector site pulled out with it. I am not that worried though, I did my first G2G transfer using fully colonized PF cakes into cracked corn in the same conditions with full success. My only problem is that I cant get the art of the casing down. Of coarse I have only tried casings three times, but have gotten contams on 2 and the other looks like it is overlayed so I scored the surface and recased with 50/50 mix that I PC'ed at 15 psi for 1 hour. I think it got overlayed because I couldn't get the temp below 80 until 3 days ago. Now I just need to get a timer for my cool-mist because the RH in the casing marth stays around 95 to 99%. Right now I have two two tier greenhouses I have set them in these tubs I found at HD they fit perfectly inside of them. I have wet perlite in the bottom mainly to make sure that the water that runs down has somewhere to go but really that has been minimal. I have a cool-mist running to each of the greenhouses and the room has a hepa filter running 247.I seem to have all the right stuff so hopefully with the temp down I will have some more success.

Hey, what's up? Fill that nipple with polyfil. Might help keep contams out. If you decide to use the baby bottle TEK again, it is good, push the nipple inside out and sand the part of the nipple where the silicon is going to go. Silicon will bond better to a rough surface than smooth.
Yes, if you don't get a 10 degree temp drop, pinning won't begin. If you left it at 80, an incubating temp., the mycelium stayed in a vegetative, or growing, state. The high humidity won't help overlay either. You should get that perlite out from the bottom of your greenhouse NOW. It's adding to your humidity problem, and, I used to use perlite in my PMP's and it always wound up coming down with some kind of growth. Red, brown, all the colors of the rainbow. Do you want that going on in the bottom of you greenhouse? Also, as your temp. comes down your humidity will increase. Good Luck.

i can't understand the actual benefits of the baby bottle tek compared to doing LCs in the same jars we use for grains

Just another way for peeps who might not access to all the luxuries we do. Saw some posts from members in Ireland and England who can't even get jars. Makes you realize how lucky some of us are.

well i'm also from europe and many things are very difficult to find (verm, lime, gauged canners, tyvek etc...), the jars sold here don't have a 2 pieces (ring+lid) top but only one piece, and i keep recycling small and big jars used for food etc, never had any need for a baby bottle though!
just for curiosity, if it is of any help for anyone, here's the way i use for filtering the one-piece lids, both for grains and LC: dig a hole on the lid, then one large or two smaller rubber tubes are placed through it and fixed with silicone, the inside end is cut slanted to make it easier for drops to drop, the tube is filled with aquarium filtering material, which is changed every time. there's no need for more holes for syringe needle because they pass through polyfill and there's no hole left when you take the needle out.

Thans for the help I finaly got the temps down to around 71 to75 by cutting the ac on 70 I will have to end up getting a swamp cooler for that room just because I don't like to leave the door to the growroom. the reason that I used the baby bottle tek is because the two times that I tried to make a LCnothing ever grew so I figured that i use the easiest one, plus I like the self healing injector site. I think its cool. but thanks for the response

yes! but agar (and you) make 2 holes for the syringe (not to create a vacuum when you suck), i make only one and i leave tubes shorter (no need of 2 holes if the needle passes through polyfill)

Only one of those tubes is for the syringe, the short one. The other is so no vacuum when drawing up syringes and some gas exchange.
