Freezing steriilization?
    #421806 - 10/11/01 09:45 AM (22 years, 5 months ago)

Another idea....sadly, i dont have the time, or the environment control to test these ideas effectivly as of yet, but trust you me i will find the area. Does anyone know the temperature ranges of most infectious strains of bacteria? because if you froze a substrate (say, whole grains), it would not only rupture the cells of the substrate making easier availability to nutrients, and kill the evil invaders on the side...opinions? Stay frosty.

im no expert by far but i do believe that freezing most mold spores and bacteria will only render them inactive for the time that they are frozen. im pretty sure once they warm back up they will just go about their way. the only way I could see this working is by a rapid and large temperature decrease such as using liquid nitrogen or possibly dry ice (solidified CO2). but putting them in the freezer will gently cool the contams into a dormant but still potentaily hazardus state. try it though, i may be wrong. i personally wouldnt waste my spores or limited supply of verm. to test it tho.

in all likelyhood it would require extreme cold temps....haven't they found bacteria living under hundreds of feet of antarctic ice? it's damn cold down there, for sure...

the biggest problem with this is that when water freezes IT EXPANDS.... in plain english, the jars will crack and/or explode if you tried this. And besides, it would have to be mighty fucking cold to sterilize by freezing.... i'm talking close to absolute zero.

Even dunking in liquid nitrogen will not kill bacteria. In fact rapid freezing helps them to survive the process. We use this method in our lab to keep bacteria stocks for years.

Two clarifications:
A: You're asuming i'm using jars....never liked the things anyways.
B: I concede the point on bacteria killing...shoulda thought first, but the first idea about the cellular breakdown of the substrate seems sound. Just throwing out ideas tho, its good to see someone else is thinking on the subject...stay frosty.

if ya froze em in any way it would prolly act like cryogenics an theyed stay dormant till they found a hospitable environment to wreack their havoc in fact i read some where that zero temps will acctually help as in preserve the various spores for long term dormancy so for keeping a vialable strain via spores cryogenics would be the way to ensure their survival .Mew

I used to move my jars to a big (-30?C) freezer right after they have been PCed and let them rest all night inside it.
Conclusions.
- I never had a single contam this way.
- Had pressure problems inside the jars after defreeze them.
- Putting hot things inside the freezer wrecks it (I didn't mind).

Peace,
MAIA
