spores on agar - the first time

ey!
so i had the opportunity to use a mates flowhood for transferring spore to agar, i personally just did peroxided agar for cloning so far, so this was something new for me.
anyways, id like to ask about a certain phenomenon which i also recognised when dealing with tissue on agar.
namely, the area where the spores (or the tissue..) starts to grow on the agar first is being concentrically colonised with a sort of 'translucent mycelium'
check these couple pics for what im talking about:


you can see the fine fibers when you hold it agains light and check the reflection, but yet it isnt white.
usually this circle grows till about 0.2inches in diameter, then starts to turn to white, healthy mycelium from the beginning and growing all the way.
i wonder what this kind of "translucent" mycelium is that appears before the "white" mycelium starts to grow, as i know its NO contam or anything, its just funky 
for all the people with more spore on agar experience, share it wit lil ohm pllls 
btw, both dishes were a week old when the pic was taken. peace ohm

and the funky thing is, it IS starting to turn white  for some reason i cannot edit my initial post b/c of some database error but as soon as its up and running again ill add daylight pics which are WAY better  peace ohm

my pink oysters (from mycelium syringe) pulled some bacterial contam looking crap similar to yours for close to a week then there was fine mycelium running from the splotches and all was well.

blimey - those daylight shots look completely different and much more promising. I hope i was wrong - get a tiny white bit transferred to a peroxidated plate asap!
this was one of my first ever agar plates:  -cyan by name and cyan by nature

why not wait till i got properly established mycelium on the dishes? like having them 3/4th colonised or so? peace ohm

update on plates 2 days later, theres quite some action going on guys 



so, besides the pfclassic plate II, they all show nice signs of growth and its soon going to be time to transfer some pieces of them to peroxided agar.
i would LOVE suggestions of what parts to transfer, as its all like uhm, pretty cottony anyway (no rhizos to my eye).
so if someone want to draw a little circe/square or something on sectors looking good, this would be greatly appreciated !
also any comments/input is greatly appreciated. peace ohm

I got your pm...and took a look at your work.
Nice work!~
In this 1st picture you can see the start of some agressive growth.

I have marked in red what to part to watch. then transfer from these area's when it has grown out a little more...to new plates. you might have to do several transfers before you get some really aggresive mycelium.

I'm realy new to everything but PFTEK. Where can I buy agar? or is it agar agar like some of the TEKs say? Is it the same as gelatin? Then I mix with dextrin or malt extract? Same thing with peroxide to get an isolated culture?

your streaking pattern sucks, just put spores in the center of the dish and that way you can pick an section to isolate when it starts racing across the plate. Here are some wood lovers. I am concerned that the top left dish might be a forign mycelium. The top right is good to go, typical ps. mycelium. The bottom is a contaminated transfer, note the mycelium is growing faster then the mold.
-------------------- Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

well, actually i did the streaking being advised by an old pro, i guess the zig zag pattern used isnt thaaaaaat bad after all 
anyway, i can clearly see your point with the spores in the middle making it a lot easier to isolate nicely aggressive strains.
i think the next time i will do it like that, but for now, i will have to work with what i got and being honest, i like the way the mycelium is developing so far.
anyway, someone, besides roadkill, got some more pointers for transfer areas? peace ohm

i love doing transfers. look at the rhizos on the lower half of this one. It was transfered from a chunk of non rhizomorphic mycelium

the problem i encountered when doing the transfers was that i didnt really have perfectly rhizo myc yet.
i transferred parts that looked the least cottony and decided to do about 2 to 3 plates of each master plate, using different areas in order to increase chance of getting a nice rhizo in one or two additional isolates.
what is going to be VERY itneresting is, that once i cloned a fruit and isolated a rhizo stem of it, i will seperatedly grow out the cloned fruit and the isolated multispore rhizo stem, i am really curious about the results 
but so far that will be some more waiting hehe. peace ohm
