Rehydrating spores?!!! With pics

I have this old pf fatass print from 7+ years ago. I made a syringe and i olny had 25% sucess rate with the spores germinating within ten days and they were taking as long as a month to germinate. I was kind of bummed about this. I was rereading TMC and i came across the spore rehydration tek. It states to to soak spores for 24 hours at 15psi 'without heat' :wink:. Since most of us dont have a way of storing something under pressure i went ahead and sterilized a pint of water with a drop of jet dry in it for 20 mins. I sucked up 2cc's of the water into a syringe and then scraped about a pea sized portion of the print into a clean petri that i wiped clean with an alcohol covered paper towel, then i waved it around and blew on it until it was dry. I squirted a few drops of water into the dusting of spores and sucked it back up. Then i squirted it into the pint jar and let it soak for a little over a week.
Whipped up some 1/2 pint brf jars like the mmgg says and knocked them up with the solution. 3 days later i had signs of life in every jar at every inoc point. No contams!! This was done in my kitchen with no gloves, no flow hood, no box. Just out in the open.

Unfucking real.  How in the hell did you not get contams with that procedure?  The spore-angel must've been looking over your shoulder and protecting your ass.  :wink:

i nver get contams doing syringes this way. I do mycelium syringes the same way off of petris. Nothing special, my house isnt super clean. Most people worry too much about contams with prints and syringes. I wouldnt do a grain to grain transfer this way though. Just spore/mycelium syrinegs

good show tahoe!!! i am sure there are still precautions that you take against contams. besides wiping with alcohol, cause just blowing on it can contaminate. cleanest breath in the west. continued good luck.

I'm just impressed.  I tried some coffee-ground added grain jars and inoculated them with a myc. syringe in a glovebox and all of them greened up on me.  I thought it was just the sterility but maybe the coffee gave the green a leg up on the myc colonization.  :shrug:

Hats off, tho, if you can pull that off.  :grin:
