Agar Work

I want to get into a bit of Agar work. I have read alot of the teks and such on the website. But a few things confuse me. Do I need a glovebox? And how do I build one? What all will I need...I don't have 100$ for a HEPA Filter and blower or whatever...Well anyways, I was looking through some asian food store today (Was bored and found some good mango shit) and saw some agar-agar. I am pretty sure I would need some of this. But what else would I need? Could I use the disposable petri dishes? And do I pour the agar into the plate, shoot some spores in and thats it? Or what? The whole concept confuses me a bit. But I am ready to go to the next stop with growing. I want to learn isolation and such. Any help is appreciated. Thank you.

I don't have any answers for you as I am just now conducting my first agar experiments but...

Thanks. I'll probably do one of those up eventually. Gotta make some more money before I put any more money into this. I gotta buy a damn car. Wrecked my eclipse :frown:

if you're in a pinch you can use your oven as a flowhood, turn on the oven and do all your work on the rack. the rising hot air helps keep microbes from falling. ive poured agar in the oven with no contams

yeah after it hardens you put one drop of spore solution in the middle

seriously though we have an entire FAQ dedicated to this if you read it you would learn alot. I highly recommened it as it has things we may miss or assume you know

I use a half pint jar with Tyvek or filter disk on top of the lid with a hole punched in it. If you use spore solution, only use a drop or two. Incubate at 86F. When the jar is colonized over, pour 1:50 H2O2:H2O solution (anything works) and scratch the surface. Suck up the solution and inject it in a spawn bag or jars or whatever. There you go - one drop of spore solution to innculate 30 jars or more, and it's very easy. Agar work done right is easy.

THanks to agar on here, I now keep my spore solution in a vacutainer and just punch through the lid for a few drops of it when I want to inoculate some half pint agar jars.

you can get a small Hepa for around 30. holmes. good investment IMO.

HEPAs aren't needed to grow bulk mushrooms, but they are nice if you have a little extra money.

I want to do like cloning and isolation for better growth. I can get the agar-agar. But what recipe is the best?

Cloning and isolation are fine, but it's also not necessary. 90% of what limits people from growing their dream tray is technique. Stuff like casing incubation and water content of substrate or casing make a much bigger difference than isolation and cloning in my opinion, especially if you haven't been growing for a long time.

Cubes grow so well that the only thing that stands between you and few pounds wet per square foot is growing technique, not some super isolate. See my avatar? That's normal yield if you get down the growing technique, and that's a multi-culture tray, all I've ever grown.

As for agar formula, I don't know if some work better than others since I've only tried the most recommended two out there, PDA and MEA:

PDA
20 grams fresh potatoes TOTALLY SKINNED or 5 grams potato flakes
20 grams agar agar
10 grams dextrose
1 liter water

Completely liquify in blender, pour, and pressure cook for 20-30 minutes. For the potatoes, you can also boil potatoes in water and use the boil water, but that's just a big hassle unless you like potatoes. If you want to do that, do a search.


MEA
20 grams light malt extract
20 grams agar agar
1 liter water

Mix with blender, pour, and pressure cook for 20-30 minutes

These work as well as any out there, and I've had great results with both. I wouldn't mess around with strange receipies. If you don't have light malt extract powder or powdered dextrose--no one does at first--get the yellow pages out, look up a beer brewer's supply store, and buy some. Or you can just order some for a few dollars online with your agar agar.

I follow pretty much the same recipies as are stated in the FAQ.

Copy and Pasted from the FAQ :

These recipes are designed for agar maintenance of Psilocybe species. All media have a near-neutral pH and should be sterilized for twenty to thirty minutes at 15 psi pressure. 500 ml of medium will pour around 20 petri dishes. Using a different medium from time to time will prevent the culture getting used to one medium.

Malt Extract Agar (MEA)
10 grams light malt extract
9 grams agar agar
500 ml potable or distilled water

Potato Dextrose (Yeast-extract) Agar (PD(Y)A)
broth from boiling 150 grams sliced potatoes
in 500ml water for 30 minutes(add water to 500ml)
(You can use 5 g of instant potato flakes instead)
9 g agar agar
7 grams dextrose( = glucose, or 10 ml honey or corn syrup )
1 gram brewers yeast or yeast-extract (optional)




If you read the PDYA one there are several different ways you can make it.

This is the one I make.

-PDYA-
500ml water
5 grams of instant potato flakes(Potato Buds Brand)
9 grams agar agar
10 ml honey (Raw natural pure honey - Beemaid Brand)
1 gram of Nutritional Yeast

I switch back and forth with the above PDYA recipy and the MEA.
And have always gotten great results.
