Rye Mycellium Syringe Tek
    #4167625 - 05/12/05 07:46 PM (11 years, 6 months ago)

OK So I know there is the poor mans syringe tek which involves using a fully colonised PF Jar to extract Mycellium from it...I tried it and hated it, due to the pf substrate clogging up my needles all the time, So here is a modified version using simmered rye.

Ok The first thing you are going to need is a 1/2pint jar. Fill this jar up halfway with simmered rye, I suggest using Masgash's Rye Tek.
I like using PolyFill in the metal lids since its cheap and works very effectivley. Now just use 2-3cc's of spore solution to inoculate this 1/2pint jar as it will take 5-6 days for full colonization. Your jar should look something like this.

Now I recommend pressure cooking your syringes pre-filled with water standing straight up so you minimise the amount of water lost during pressure cooking for 30mins @15psi. If your PC is not large enough to handle your syringes you can always pressure cook them empty and then pressure cook a jar of water as well. I prefer not to do this as it just adds one more step to the Syringe making process.

REMEMBER TO ALWAYS FLAME YOUR NEEDLES WHENEVER YOU ARE PUTTING THEM INTO THE JARS LIKE THIS.

The Next step is to remove the polyfill from the lid of the jar and take one of your syringes and inject the water directly onto the rye inside the jar HARD, like so.

Now there is always a little bit of water that gets eaten by the jar, so eaither pressure cook extra syringes or just be prepared to not get a full syringe back( i mean like 2-3cc's less then what you started with). Shake the jar SLIGHTLY (Swish it around a bit in a circular motion) to break off some of the mycellium from the rye cake..but do NOT break up the cake itself as this will cause kernels to clog your needle. You must tip the jar on its side and slid the needle into the hole in the lid at as steep an angle as possible, like this.

Then just suck the water out of the jar and VOILA. Instant Mycellium syringe. You can easily get 500cc's of liquid Mycellim out of this jar proved you have enough syringes of course. An added bonus for you karo lovers out there is to pc a syringe full of karo water also and use this tek the same way, but then you will have a syringe in which the mycellium will actually live and grow inside. I have used plain water syringes up to a month after they have been made with no ill effects though. I really hope this helps anyone who wants to make their spore syringe they paid so much for go as far as possible. Its also a very easy way to spot contams that may be harder to spot growing spores out in karo.

Peace

ok, why not use the mmgg's boiling distilled water and sucking into syringes?

I prefer to be extra sterile and enjoy the peace of mind that comes from PC'ing syringes. I would imagine you could use the boiling water method also though.

why not 1/4 full i think 1/4 full jars would do alot more damage to the mycelium getting sprayed HARD. does 1/2 full jars produce more syringes?

would wide mouth produce more syringes in combination with the 1/4 or 1/2 fullness?

1/4 Pints would work also, and probably colonise faster, but I would imagine the 1/2 pints would produce more inoculant and also I didnt have any 1/4pints.

Interesting tek, though what would be the benefit compared to, say, innoculating spores into a karo/distilled water/potato flakes solution. This is the method I was taught by someone in the IRC chat, PF jars are unecessary.

Is it perhaps colonization time in the rye? My karo jars took around 15 days for a thick mycelium buildup.

You nailed it on the head, speed and also amount, like i said you can have 500ml of liquid myc soluction in 5-6 days whereas with karo you have to wait a hell of a lot longer to get that amount. NOt to mention as i said in the post, very easy to spot problem syringes.

I tried this with a 20ml syringe and a quart jar a few weeks ago just for the hell of it, didnt go so hot. Only got 3 cc's back of the 20 (used a quart that had been colonized for a a few weeks) and then...it contamed. I think it was the batch though, I was in a rush for PCing....

Good tek though! I'm sure if its done right...

ya what greg said, watching karo for contams isnt like watching a jar for contams. karo contams can sneak in @ the poly. the cobweb can also look like mycelium in liquid karo also.

Dear GregHimself:

Q: I have leftover PFTEK materials, could I use this for the "1/2 pint" jar just as easy instead of rye?

YOu used quarts Scat thats your problem..as for the contams i dunno, i have never ever gotten contamed from a clean grown jar of rye.

The problem with doing this tek with PF materials is a. the cake will absorb WAAAY to much water once its injected, AND the BRF and VERM mix just clogs up the needle so its near impossible to suck what little water is left back into the syringe.

Yeah, I know where I had a bunch of problems...I was just stating my failures because people read things and try things that are "close" but not following the tek to a "T". I wasnt following a tek...I was just seeing what would happen..the quart went to h-poo and gave me lots of shrooms, and the jars were extra..no loss to me, may be to someone who tries what I did though.

Basically point is, follow it to the "T"!
