Agar transfer

when doing an agar to agar transfer to isolate a single strand. when you cut out the chunk of agar should youplace it on the petri dish mycellium facing away from the new agar medium or sandwich it with the agar? i sandwiched it basically and instead of growin out it is growing through the initial wedge first.

When making a transfer, I take a piece of mycelium no larger than a grain of rice, then place it myclium up on the new plate. It will grow down the sides of the original piece of agar, then into the new dish. An alternative method is to not take any agar from the first dish at all. Simply use your scalpel or inoculating loop to scrape a small piece of mycelium, then deposit it on the new dish. If you're doing strain isolations, be sure to take as small a piece as you possibly can.

you will also notice that the mycelium is growing in and on top of the agar. When you cut out a section you will notice the mycelium that is inside the agar will recover first and there will be wispy strands coming out the sides.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.
