newbie needle question
    #4147500 - 05/07/05 05:57 PM (13 years, 1 month ago)

Ive got some jars that are now drying and Ill bee inoculating them later tonight.
I wondering about sterilizing the needle, this will be my first attempt and with somewhat limited resource-

I will be using a bic lighter and plan to wipe the needle after heating with a paper towel and 99% isopropyl

will this be safe?

many thanks

That's fine, but flame sterilization isn't needed. For needles and inoculation loops, a quick wipe in 70% isopropyl has never let me down. 70% evaporates slower than 99%, so it's a better buy than 99%. Just a note for your future reference.

drying!!??

I meant cooling..

.

wow thanks for the swift responses

yeah it seems 70% is a better way to go, but I rushed to get 99% before I saw the additions in the sterile tek-

thanks again

I flame sterilize between each jar, and wipe with 70% alcohol between each hole.  This might be overkill, but I would rather be safe than sorry.  Good luck

Using my portable range, after each jar, I clean the needle with alcohol wipes, then hold over the range for a bit. I don't like the fact that lighters leave the soot on the needle. An alcohol flame would be better.

holding the needle in the blue portion or about 3/8 inch above the flame will prevent soot buildup with butane lighters
