Yeah but out of a 1000 ml solution, every bit of it should be useful. In other words, make sure your swirling your solution well in order to disperse the mycelium accordingly.

O.K. I may be a complete dunderhead, but the argument in this thread confused me.. Is mycelium like spores, in that it can end up being micsroscopic chunks that will colonize jars or spawn? Or are you supposed to get a hefty amt. of visible mycelium in each syringe? Or,are you just supposed to shake it with a marble or two long enough so that it's evenly distributed in the syringes??

you *could* use the marble, and that would provide a more even mixture of mycellium to solution, or if you want to give your jars hefty kick in the ass, let the strands grow out and suck it into the syringe selectivly. i prefer the latter cause i dont work on a large scale and timing is everything, but for large grows, the marble might be the best choice.

Marble's take up to much space IMO...

I just shake the shit out of it for a mintue...The myc. gets shaken up Very nicely...

But if I had to shake more than 5 jars for more than a mintue...i'd probably go insane :smirk:

What kind of marble are you using Road? hehe, I can get 5 marbles in a 12cc syringe. I only have a couple jars of LC going at a time, they go a long ways. And you can grain to grain from that.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

Roadkill, when you started your solution, what did you use, spores or mycelium? How much? How long do you shake the jar per day?

Agar, I also have light malt powder and dextrose. How much of each do you recommend for your dextrose/malt mix? You seem pretty keen on automatic stirring. Does this speed up the solution colonization even further? What do you start your liquid cultures with, mycelium or spores? If mycelium what is the source?

Spores

1cc to 2cc

I just twirled them after inoculating...then a couple days later.

that was it.

Sounds real tempting, Roadkill...

but then I looked at my half pint agar jars that have been incubated for about 3.5 days now each with only a single drop of spore solution out of a 1/2cc 29 guage insulin needle. The surfaces are about 30-40% colonized. Using a very small needle like that, a single half CC of spore solution can inoculate 20 to 40 jars of agar surface! And then each jar can be blended with 200 ml of sterile water to inoculate a lot of grain. One drip of spore solution to a 3/4th covered agar surface, enough to inoculate pounds of grain, in about a week. What's more is that on agar, I have the flexibility to see contamination easier and I can see which surfaces are showing stronger and faster mycelium growth.

I am not saying that one technique is necessarily better than the other since I've never tried the Karo technique, but it does show that there is more than one way to skin a cat.

By the way, a 29-gauge needle is so thin that you barely can feel it going into the skin. It's about 290 um in diameter (less than the width of a human hair), even less in the middle where the liquid flows. I was amazed at how little spore solution is needed to inoculate agar and how tiny the spores really are!
