So If Im doing my math correct here... In my half gallon jar I should only need to use 2cc's of the syrgine?(.5cc per half pint, two pints in a half gallon) Thanks for the info on the agar.... thats a wonderfull indea and my mother does have a pressure cooker i can borrow.... so i think i will make bottles up with remainder of the syringe..... Also after doing much reading I am going to go with the perlite tek and will use tops of butter containers to put my crumbled cake ontop of the perlite.... Hmmm I think my plan is comming together:) Thanks everyone... I can't wait to get started.... Yoda

I love this tek. Thanks for bringing it to light Agar. I knocked some bottles on 06/15 using this tek and saw mycelium forming on 06/19. That's only a total of 4 days. Obviously it'll be weeks before it's really thick, but I'm on my way.
I used 1/2 CC per bottle. That's it. The rest of the syringe is stored in a vacutainer in the fridge.

Okay so if one fills the nipple with silicon to keep "air and water tight"...does this mean LC's don't need air exchange?

I prefer some gas exchange. But, sealed - you will still get myc - just not as thick.

If Im using agar (no pun intended) to make the LC and not a syringe, will I maintain a single genome during the incubation? The myco wont reproduce different genome type strands, right?

ticktock said: My 2 cent additional suggestion; Do everything just as described above. After you see the first development of mycelium, inject a volume of 3% peroxide equal to 1/20 the volume of the liquid culture. Within a minute, the mycelium will head for the surface and float. They have caught tiny bubbles of oxygen and will use it up in the next few days. Repeat when they sink again. There you have aeration without the need for super filtered air. ticktock
Anyone have a reason this wouldn't work?

Don't baby bottles have air exchanges in the caps? So that a vaccum isn't formed by the baby sucking one down? Apparently the ones for cattle do. My friend decided to scale your tek up. He bought like a 1.5 gallon COW baby bottle and turned it into an LC.
