Preserving for future cloning..
    #413610 - 10/04/01 11:25 AM (23 years, 2 months ago)

I need to stop growing for a few weeks as I will be moving to another place..

The thing is, I have some niiice ecuadors fruiting and there are a few beautiful specimens I wish to clone from in the future.
I have heard about the possibility of extracting some inner tissue and storing it in distilled water for future culturing.. will this work?
If so, aside from watching the temp of the water, what other factors may I need to take into account for this venture?

Any and all help will be appreicated.

peace and thanks,
uniboner

i tried doing it recently. i had some cut up inner stalk in a jar of water and put a big piece on some agar. i got no growth though, but i may have made the agar too strong as in over 4% malt since it looks dark and i didnt bother measuring properly. ill try it agin in the next few days and keep u posted

It could/should/theoretically be OK. . .more often this technique is done with agar growth, thus ensuring contam-free dH2O. Remember that if you get a tiny unseen bacteria or mold colony in your liquid inoculum, the subsequent expansion will be disastrous.

If your fruiting chamber is pretty clean, and your transfering technique is good, then putting a chunk [skillfully extracted] into dH2O will maintain the strain for a long time, esp. if frigidated [I think it was Anno that pointed out a reference claiming 2+ years without loss of vigor]. I can't speak for peroxide with such a tech, some fish antibiotics might help.

This has worked for me. Nine parts H20 to one part (3%) H202.

you could put some stripes of inner stem matter in honeywater, by the time you have moved you'll have mycelium clouds floating around... try to avoid big pieces, they're likely to rot... thin stripe pieces will do the job... you've got time.

i hate witty sigs / if anything i say has something in common with reality, this is a merely a coincidence.

Thank you all for your help.
I would be a much more frustrated amatuer mycologist without your tips.
