Tape covering innoc holes

Should I sterilize tin foil squares placed over the taped holes? So I can put them back over the jars after I puncture the tape w/ the needle? Or is this silly as when I take the foil off it will be exposed to said airborne contams I am obsessing about?

Don't bother recovering them with foil. Just make sure you've got a layer of Verm on top of your Cake, which helps keep out contams. And don't poke through the tape with your syringe either. I've had problems with a freshly flamed needle melting tape, and other problems where the adhesive gunked up the needle. So lift back the tape and inoc. You're just going to remove it later. The jars will halt their growth if you leave them on and they can't breathe.

How soon after inoculation should the tape be romved?

This might be superstition on my part, but I wait until a good portion of the substrate is colonized, like about 30%. If you've got a verm layer, you could leave it off immediately after inoculation.

I will have a verm layer, but superstitution sounds good for me, that is if your fairly sure that the lack of air exchange won't hurt anything?

i taped the holes on my jars because i noted that magash had done it in his PFtek pictorial. right after i inocculated though i saw that most people suggested not doing it, and that magash explained that he did it only until he saw myc growth. i took it off after like day 3 of incubation and after that they took off. i also turned my jars upside down, which also helped it seems.

i don't think i'll tape again, it was just a pain in the ass to remove in my glovebox

I have no glovebox, I'm relying on airborne Lysol.. So I think tape wouldn't hurt...

It won't hurt any growth for the first few days of colonization. You'll be fine. I turn mine upside down as well, usually during the last 10% when it's having trouble getting to the bottom.

My 10 jars are infested by green mold, right now. It's why I'm concerned.

Is this tek okay to prevent bacterial infection during the inoculation ?

Once the jars are sterile and cool, one by one you can inoculate it and put tape (sterile) on each hole.
**Don't put tape just after the PC'ing because when cooling, the jars will contract a bit (I was having plastic jars) and some bad air will be aspired inside the jars during the inoculation (what's happened to me).**
After 3 days (or 30% of colonization), remove the tape.

I think you're supposed to PC with the tape on.. I sterilized (steamed, I'm doing the PF Tek) w/ the tape on. I took the tape off to innoc, and left it off. I only innoc'd yesterday, so I can't tell you if I'm succesfull yet (crosses fingers)!

you ever try to pull tape off your lids after its been PC'd? its a bitch. the best advice as far as that goes is to use a folded over piece of foil in place of a lid, remembering to screw the foil down with the lid ring. no messy tape gunkin up the lid or the needle, no large hole size or guessing where your small holes are, just puncture and cap with another piece of foil.

or hey! put tin foil over it and p.c it and then take to glove box or sterilse area and pull tin foil off and shoot it up and put tvek over it...its free

I recommend using no tape just foil, it's never failed me and I've also never had a stalled jar either! :wink:
