13 days no sign of growth!
    #4096807 - 04/25/05 09:37 PM (12 years, 6 months ago)

Alright I have a dilemma; it seems my Mycelium won?t grow. I have prepared 12 jars using the PF method and I will go into further detail as to help isolate the problem and hopefully find a solution.

Here is a list of materials and where they were purchased from.

Item and description Where purchased

10cc Syringe of Psilocybe Cubensis Huautla Sporworks
10 sterile 10cc Syringes Sporeworks
1 8 quart Bag of Whitney Farms Vermiculite Coarse Ace-Hardware
1 80 oz. Bag of Californian Brown Rice Grocery Store
1 Dozen Wide mouth/sholderless half pint Ker?s mason/canning Jars Wal*Mart
1 23 quart Presto Pressure Cooker (Stolen) Ace-Hardware
1 Very nice 90 Gallon Fish Tank Heater (Stolen) Pet Store
1 48 Gallon Rubbermaid bin Wal*Mart
1 Digital Thermometer/Humidity gauge (Stolen) Wal*Mart

Alright so my procedure for the sterilization of the 12 jars.

The jars remained in packaging up until sterilization, I then removed them and poked 4 holes in each jar lid.

I then mixed the substrate as follows, 3 cups normal tap water, 3 cups Brown Rice Flour, 6 Cups Vermiculite. I also added ? of soy protein as to give them as much food as possible. I wanted to make sure there would be enough water so what I did was completely soaked the mixture until there was just a little puddle of standing water I mixed and discarded the excess water.

I then gently spooned the mixture into the 12 jars making sure there was plenty of free space for air. I then sealed the holes on the lids with Masking Tape and secured jar lids.
I placed all 12 jars in the pressure cooker with the correct amount of water and let it cook for 1 hour at 15psi.

I let the jars cool over night in the PC and then removed them and prepared for inoculation. I sterilized the needle with a butane torch and cleaned it with alcohol and let it cool. I then inoculated 12 jars with the 10cc syringe; I thought I could stretch it. There was actually more like 11 to 11.5cc in the Spore Syringe so it worked out great.

After inoculating I then resealed the jars and set them in the sterile incubator. The incubator is a T.I.T. (Tank in Tank) using a large 48 Gal. Rubbermaid, A fish tank heater, and the P.C. (Pressure Cooker) set inside with the jars. It is filled up with enough water to be as high on the sides of the P.C. as it can without making it float. The fish tank heater is set to 86 degrees Fahrenheit. I know the temperature and humidity because I have a nice digital Thermometer/Humidity gauge, It reads inside and out side temperature as in inside the sealed P.C. heat and humidity and outside. The P.C. is actually totally sealed inside the Rubbermaid as to keep out contamination and keep the humidity high (which I have heard is good but I don?t see why the jars are sealed).

I have checked the jars every day since I started incubating, which is 13 days now. I have not seen any sign of Mycelium growth nor contamination, there is simply nothing happening..

At first I thought that maybe I should try leaving the jars unsealed.. So after about day 8 I opened two jars every day and allowed fresh air exchange. Since I have been doing this there has been no contamination but still no growth.

I now think that there may not be enough water, although the cakes looks moist and there are no puddles or drops of water perhaps there is not enough.

I know this is a lot to read and I know this is only 13 days but I would really enjoy either some sign of growth, contamination, or feedback from the great people here.

I am open for further questions and or comments?

Thank you, a fellow shroomer and wanna be mycologist..

What is the relevance of the 10 sterile 10cc syringes from Sporeworks? What were those used for? I hate to say it, but it looks like you may have a bad syringe of spores. I can't really see anything else that you did wrong.

Anyone else?

I have the same problem, I ordered huatla spores to, they really are a pain in the ass. 3 of my jars refused to show any meycelium growth, while others showed a small speck of mycelium that never grew any bigger than about the size of a dime. So what I did was I took a jar prepared the substrate accordingly, and knocked up each point in the jars with 1cc of spores. That did the trick, now im going to birth it and the first shroom I get, I'm going to use to make a spore print. Fresh spores, and I'll knock up 10 jars with the thickest solution of spores ever.

I'd ask for a refund...  And not write such a convincing argument for the prosecution as it appears you're in the US

The thing is huatla spores arent very popular for some reason, I think that those spores just sit in storage for months at a time until they get old, then when someone finally buys them, they're just bunk.

I've bought from Lil' Shop of Spores and SporesDirect. I have some Puerto Ricans going from SporesDirect (Buy one get one as a member) and other strains going from Lil' Shop of Spores. The strains going from Lil' Shop of Spores are kicking ass, while the other spores are colonizing very slowly.

It might just be a luck of the draw though.

1) Why are you adding Soy for protien, I have never heard of this?

2) Why are you adding extra water? Trust me they are not lying when they say a ratio of 2:1:1

3) Why did you open your jars? This is just asking for trouble.

I have also found that sometimes spores can take a while to germinate. I recently had one jar that did nothing for like 20 days then exploded with growth. Yes, it is true germinations will usually occur around 1 week, but that is not always the case.

If this is your first time growing, I would suggest not experimenting with things and following the tek to the T.

I would suggest getting a new syring and starting over, as it may have been a bad syringe or maybe just bad technique.

You do seem like you have thought about this alot and put much effort into so Im sure will suceed soon! Just keep trying.

Perhaps I will ask for a refund but I doubt they will give it to me.. Until then are there any suggestions on what to do with this mess? I was thinking of putting all of the substrate together outside as it is the right time of year here for Cubies.

I guess the 10 sterile syringes are to show that I would like to know if anyone would do a trade.. Perhaps 2 or 3 empties for 1 in return full of some spores..

Also I am not in the US I am in Canada!

This is my first time growing.. I have done a lot of reading and I know I should not experiment my first time but eh.. what the hell.. Soy protein.. I figure it has a lot of nutrients and good stuff for growth.

I guess I will hold out another week or two and see what happens!

Thanks for all of the replies I was not expecting so many so fast! That is why I love Shroomery, it is a nice crowd that usually knows what they are talking about!

The syringes are most likely fine.

Dont use brown rice, use birdfeed. I've had the exact same problem, in fact, the mycelium only started to grow, and in very small amounts on like the 25th day. The thing is, some farmers put fungucide in it to keep it from contaminating with fungi, and well, therein lies your problem. Go to your nearest produce mart and get some finch birdseed, will work like a charm.

Good luck!

it happens that you do get bad syringes. i think a foaf has come across two z strain. time and effort have been waisted and money if if the seller wont replace them. If your not getting any growth contamination or otherwise you have a sterile environment. If you dont have too much or too little moisture then it is your spores.
i'd suggest not checking your jars for a week. make sure the tape is off the tops of your jars and layer a sheet of foil accross the top. Its possible to love your jars to death. if you dont see grwoth in a week let it go another week. make sure you are incubating at 82 to 86F. if and when you see contamination toss your jars and try again.
