Before i give this a try has anyone tried subsituteing jello for agar? Not the flavored kind, just the gelatin that can be bought from the grocery store. If agar is used for its sterile/pouring/non nutrient ease i dont think there is much of a difference. Agar is just the vegetarian version. Of course some nutrient would be added ME or PD or somehtin. ANyways i figured i am not the first so i should get feed back

Agar is preferred over gelatin because gelatin can be digested by a wide variety of microorganisms and fungi, whereas agar can not.

but for the peron who doesn't want to drop 20 dollars just to try somthing they end up not likeing. If the only problem is a wide range like agar be extra sterile. Also once you add the malt extract/ PD/ Dextrose arent you kinda nullifying the diversity thing. Or does agar have special enzymes which block other things completley

I pay under two dollars for enough agar to make 50+ dishes. I use raw agar agar from an Asian market.
The problem isn't sterility in the case of the home cultivator. Agar has chemical bonds in its structure that are not easily broken down by natural enzymes, whereas gelatin has bonds that are. If you use jello you run the risk of ending up with cultured goop. If you use agar, the cultur will grow only on the surface which is more desireable for cultures.

thats the answer i was looking for.
now about this agar from the asian market, is there anything special to be on the look out for? Or is all there agar the same?

Make sure to get agar agar. It comes in powder and in raw form. I buy the raw form which looks like long strips of clear seaweed in a plastic bag.

Couldn't you make a poor mans cloning agar using H2O2 and jello? If your conditions are sterile it seems plausible. This would be in the instance that someone has a box of jello and can't find a cheap source of agar locally.

cheap Agar is everywhere.Health food stores,Asian markets,Arab markets,the corner bodega,high end grocery stores and major grocery stores with foreign specialty items and ingredients,etc...many places,cheap,just look around.gelatin can be used but doesn't work nearly as well as agar,and since agar is so cheap,nobody bothers wasting time with gelatin at the chance you may have problems or end up with "goop" as Joshua puts it.

I am surprised vendors don't offer small amounts as freebies, it costs very little in bulk and they would pre-add peptone or other nutrients.
Anyways, I always thought agar was preferred as it is a solid at incubation temps, AFAIK gelatin is liquified at 25C.

I think the melting temp is/can be higher, but that is a good reason as well.
H2O2 might help when added to gelatin...I have no experience with that though.
Agar is so easy to find and has many benefits so I really haven't experimented with gelatin.

Also, you can do an even cheaper and easier agar substitute with BRF paste. There should be plenty of info if you search, but basically you just mix BRF and water into a thick paste, spread it in the bottom of a 1/2 pint and treat it just like it's agar. I know it's not a perfect substitute, but it does the job well.

I think you are right, as you can buy a brick of "undiluted" jello in the shops. You must need a high concentration to keep it stable though. People have used cardboard soaked in nutrients too.

jell-o is too rich, nute-wise, for a good agar substitute, plus it'll liquify over time.

I've used unflavored gelatine and beef bullion to make an agar substitute it works but as you mntioned, it breaks down rapidly to a liquid, usualy in 3 days I have growth and it's halfway to a liquid state, I usualy transfer it at that point
overall, gelatine/jello sucks

LOL! I was hard up for petri medium for a mold test and decided to use Jello in the place of MEA or exc. Cubensis spores will germinate and grow out on Jello, but it wants to liquify when the temps reach 75 or so. It's rather useless really for an agar substitute, but it worked for telling me how dirty the air was in this particular room....
