liquid culture question

how much myc do u need in the jars for it to be use full, afoaf had some little jars with hardly any myc floating around, theres a lil bit in there like a buncha quarter pea size specks floating around, if he sucks what he can up in a syringe will it be enough to knock up a buncha jars. please help

hey agar--is there a good liquid culture tek out there that you would recommend? what is the advantages of doing liquid culture? fast colonization? i'm a bit doubtful, just because it would, for me, add several more vectors of contamination, and that always worries me.

Liquid culture do add lots of risk for contamination, but they increase colonization time outrageously. Its well worth doing, once you perfect your methods contamination won't be a problem either. Lipstick mold seems to love liquid culture, and at times it can be nearly impossible to detect. Good luck, I suggest giving it a try.

about how long after inocculation are those jars Agar?

They are around a week old. Mostly started from an agar isolate wedge, or tissue, or single fully colonized WBS grain. Spores take a few days longer (germination time).

You want to work under a flow hood, in a glove box, or if you have neither - on a very sanitized bathroom counter, with mask, hair cover & gloves, with slow calm movements.

2% dry dextrose 2% light dry malt.
Mix, pour in large jar, PC 20 minutes @ 15 psi.
Remove - cool - pour through double coffee filters.
(removes any gunk, or carmelization that forms)
Pour into individual jars & PC again 20 minutes @ 15 psi.

Lids as below make it very easy to keep it sterile.
Just rubber tubing, RTV high temp silicone gasket maker & polyfil.
(you can do the same with a metal lid)





PS.... you can get good small bore rubber tubing at any auto parts store (vacuum hose for windsheild wipers)for cheap.

agar prec the info, tried a couple of times with liquid culture but always contamed. great pics helped out thanxs again. is it necessary to use the dextrose? the tek i used said only light malt at 4%. too much?

4% is the max sugar solution you want to use. Any more becomes TOXIC to myc.

Light dry malt alone works. I use dry dextrose & light dry malt @ 2% each - because I have had great results with it. Hell, karo works.

i did the microwave tek i saw on here and used 1 Teaspoon of karo in 5 ounces of water. Is this too much sugar for the myc.? It seems to be growing decent now after 6 days at only 70 degrees.

Exactly or approximately, how much mycelium should be in the container before it is good to put in a syringe? Mine has been incubating for 5 days and if I hold it up to the light I can see tiny strands of what looks like snot or a lugie. Probably the right stuff just doesn't look like the pics I've seen. And I know be patient, I just was curious how much was necessary.

I find that when using spores in a malt(or whatever) culture that since the spores sink, %90 of the time my first mycelium balls are formed, and covered in the sugary sediment. As long as it has not been carmelized I have found the sediment beneficial.

Besides, since its best to mix the malt and distilled/spring water under very low heat, why not just filter before the PC? :confused: :stoned:

i have light malt extract, Agar agar, instant potatoes and honey some say 7 gr malt extract to 200ml others say 1tbl per cup none use agar can you help, using powder light malt extract. Im not getting conatm but no culture either. I tried 1 tbl malt to 1 cup water with nothing else what am i missing here

This is my favorite recipe
20g Dry Malt Extract, or 30g (liquid)Malt Extract @1000ml Water, for LC.
20g Dry Malt Extract, or 30g (liquid)Malt Extract and 25g Agar @1000ml Water for petri dishes.
No Dextrose, potatoe or honey but you can (optional) add 2-3g Dry Yeast @1000ml for additional nutrients.
Thats it.....
If your spores dont germinate on this LC then they are maybe dead (too old or wrong storage).

There are vitamins and other nutrients into the yeast.
The myc will benefit from them.

spores new from sporedepot in fridge for 5 days per haps not right forum but new to forum just came back to the world after five years of solitude in northren woods, is the sporedepot reliable
