Hello, I was hoping that someone with experiance with agar could give me their oppinion.
My friend has read the mushroom cultivator along with hundreds of other documents and decided to try agar.
He poored 17 dishes in a glove box (the box doesen't have gloves, just holes). Once cool he innoculated 5 dishes with a eq print from ralph. he used disposable inocc loops (package sterile untell opened).
When he was done with that he wraped all 17 dishes with strips of 2" siran.
all was done in a simi sterile enviroment ie clorox disinfectant every where including in glovebox.
any way 3 of 5 grew large amounts of green mold in the center of each dish.
Question.
Were the contams from the sporeprint or from the air.

I guess you sterilized the agar for 30 min at 15 psi? If so and you used good sterile technique then it may have been the spores. You never know with aspirgillus. I got a contaminated pes amazonian from the fsr. Oh well...It was from 5 years back. I have only done about 5 plates of it and not one spore germinated on MYA. It happens. Any one have luck with his Wild cyans yet?

Jahson, with older spore prints, I usually have much better luck if they are allowed to rehydrate in a small amount of sterile water for 24 hrs. before inoculating. Since your working with agar, a pipette can be used to transfer a couple drops of spore water onto the agar.

If possible, repeat the agar tek u used but dont innoc the plates. Let them sit for a few days and any contams will show up fairly quick - this will give you an indication of how sterile you are being. Get some gloves if possible stops skin flakes falling onto the agar.

thanx for the replies i transfered the most promising sectons from the 2 clean plates to 5 new plates i guess it was the print.

It's most likely either from the spores or the tool you used to scrape the spores off the print and put it onto the dish. To determine which one it is, sterilize the tool completely with isopropyl alcohol and flame the tip before use.

I used disposable plastic loops so i could not flame sterilize and i was under the impression that alcohol distroys spores.
Does alcohol distroy spores?
