liquid culture timing

I was bored the other day so i fudged a microwave tek liqiud culture. I used a pint jar with saran wrap on top then capped it quickly when it was done. I used about a cup of water and a splash of light karo. How long does it usually take to see myc growth. I dont have an incubator but my temps are in the mid 70's

how many cc's of spore did you put in it?

Your screwed! The cap of the jar will harbor contaminates. That's why pc'ing is essential for this tek, even a steam sterilization works nice.

will to much karo hurt it, cuz i think i put a bit to much in.

lets say contams are not evident here... i have never had problems in the past. How long should it take, considering i didn't put to much karo in.

Don't know what to tell ya. I use 3 tablespoons of Karo per 1/3 of a qt of water.

Wow hot, thats a ton more then everyone suggests. People told me I was crazy using 2 tablespoons in a pint...

To much karo, its been shown does hinder growth...you want 4-5% I guess. I think I use closer to 10% myself, I get growth...not as thick as some others but I havnt done a culture in a while...about time again I guess.

a mushroom is 92%-96% water, the rest is just solidified mycelia. the sugar/carbohydrates in the honey/Karo are just nutrients. the mycelia just needs that LITTLE bit of sugar and the 96% of water to grow. when you throw the balance off it has more nutrients than water and doesn't have optimum growth parameters.

Hmmm. I'm not a math wizard on liquids LOL. I'm not sure how much 1/3 of a qt is versus 1 pints. There's 2 pints in a qt right? Uhhhhh. Heck if I know! I've never had a prob with Karo and these measurements though. They grow optimal! Honey on the other hand is a different story.

Two pints in a quart, yes.

Those are just like mine....not as thick as agars or some others I've seen...thats what makes me want to try out less sugar (and something other then karo). And agar gets quicker colonization with LC's then grain to grain transfers...I can't do that...growth with LC's is quick compared to spores, but not G2G.. I'd suggest trying out a little less (maybe 1 tablespoon to that amount) for a culture. Use your regular recipe and try to start from a tissue sample instead of MSI. See if theres a difference. This is what I'll be doing anyway..just found some kick ass LC bottles too yesterday..gotta see if they PC today..

Hmmm. That's interesting. Yeah, Agar's cultures are nice. I'll proly use less then. Any hints on the new LC jars? :cool: What about nutritional yeast in LC's? I think i've seen Agar use it before. Not sure though. I've definately seen his juice/beer cultures and they look nice.  :thumbup:

Definitely stick below 5%. Remember when microwaving you will loose water and concentrate the sugars so what started at 4% will be 8% if you boil off half the water. Don't be scared of putting large metal lids in the microwave either, only tinfoil or metal on its own in a microwave will cause trouble.
Too much sugar will kill it instantly but too little will only make it not grow as much (should be about the same speed growing)
I make all my liquid innoculations in the microwave, also put in some broken glass, so when you shake it it cuts up the balls of mycelium so they are easily sucked into the syringe

Weight in grams (metric), Volume in pints (Imperial) - why not just stick to metric, so much less fiddlier :smile:

10g dextrose, 250mL distilled water, 10/250 = 4% Simple!!!

(don't get me started on doses in metric grams, while the most famous growing guide is in imperial 1/2 pints!!!)

I'll be trying a teaspoon, a tablespoon, and 2 tablespoons per pint (1/2 quart). I usually use the last one, but I think the first one (teaspoon) is going to give the best results. I'll be trying agars recipe from that link though when I can fine the malt and dextrose...

OK. Let me go ahead and drive the rest of the nails in the coffin then. 250mills is equivelant to 1/2 pint, right? If not, please correct me! Never paid too much attention to the math for some reason. Thick scull proly! :smile:
