Very interesting agar contamination
    #4020613 - 04/06/05 09:30 AM (11 years, 7 months ago)

After pouring my agar, I let it sit for a few days to check for any contaminants and, of course, I always get some. This time something unusual appeared in one of them. Check it out:

Anyone have any idea what could this be?

Also, after doing a transfer, something appearing to be bacteria showed up in one of my plates:

I guess there isn't much way of savaging this ones...

Any comments well appreciated

Junka

But it's not green at all, has never been... since it first appeared it was rubber like black thingy in the middle and a bunch of gray hair around it.

The other one is bacteria for sure...

The bacteria is most likely either from your fingernails(wear gloves) or your breath(use surgical mask). The top pics are of an imperfect fungi of some type. You shouldn't be getting any contamination. Use a glovebox if no flowhood, and only remove the lid long enough to pour the agar, then put the lid right back down. Hold the lid in your other hand while you pour. Wrap with parafilm immediately. Glass petri dishes have a horible seal. I tossed all mine out.

It's impossible to ID just about any mold from a pic, but I'd lean towards Mucor or Rhizopus spp. . .they both are very fast-growing, sometimes can get beefy enough to lift the petri lids! [yuck] Could be just about anything, though; The solution is as described; Fresh ziplock bags can help ward off airborne problems similar to parafilm, but make sure the temperatures are equalized before sealing to reduce condensation problems.
