Does anyone who is experienced with mushroom cultivation know if it's ok to cultivate edible mushrooms such as portobello and "others" in perlite as opposed to the traditional use of vermiculite?

Perlite is used as a humidity source to induce pinning, vermiculite is used for water retention in the casing layer, and sometimes the substrate itself. Two different applications here--you need to specify.

Are you asking if perlite can be used to humidify these other species? Yes, it can.

I was wanting to know if it could be used in the substrate along with rice flour.

Well screw. I had no access to vermiculite at the time I was cultivaating. I just tried using perlite in 3 jars. Not that many but still. I guess I'll have to go pick up some vermiculite tomorrow. Just out of curiosity why wouldn't perlite work. I'm a novice so please excuse my ignorance.

another question pertaining to perlite...
how often do you change it when colonizing a casing/fruiting

I'm not sure. Perlite has great water retention when used in potting soils, but the texture doesn't fly for mushroom substrates.
