Kronny's latest experiments
    #3969152 - 03/25/05 04:16 PM (11 years, 8 months ago)

Yea so on 'some other' forum I read about someone sterilizing jars of WBS in the microwave, so I decided to give it a try, as well as a jar in the oven.

He said to put a jar of WBS in the microwave heating for a minute in a half for 1min 30seconds at a time, I did it 5 times. This was a quart sized jar, covered with a layer of tyvek and I added a little verm to the mixture.

Oven jar is a pint sized jar of wbs+verm, with a layer of tyvek and a layer of foil (one reason I'm likin the oven).

I baked the jar for about 40 min total at 250F. High enuff temp to sterilize but not high enough to burn, in a short amount of time in the oven anyway.

Innoc'd microwave jar with PF myc grown in sterilized rye water, oven jar with PB myc grown in rye water.

3 days later, nice myc growth and green meanies in the microwave jar. Oven jar, light myc growth, no contams!

5 days later (today) Microwave jar heavy myc growth as well as green meanies, it appears contam came from myc solution, I was suspect of PF syringe from the start because it was homemade.  More myc growth in oven jar, slow but progressing (I think PB colonizes slow), also I don't feel like turning on my incubator for 1 pint jar.

So things are looking good for the Oven tek workin out, and I have to give the microwave tek another try especially in a smaller jar with sterile solution.

:syringe:

I'm assuming that you soaked and simmered the wbs first. That swells the grain to it's capacity to hold water.

Pressure cooker; Raises the temperature to about 250F without boiling the contents of the jars. All of the water that starts in the jar stays there.

Oven; You got your jar to 212F. At that point the water began to boil out. The temperature may have gotten a bit higher than that, but at the expense of losing water. You will only get up to 250 when all the water is gone. What's missing here is the pressure. Without that you can't get it hot enough and keep all the water.

Microwave; Same as above, but the grain will have boiled from the inside. I suspect that the grains have burst.

There's a reason why we use a pressure cooker. It one sure way to kill everything without drying it out. Feel free to experiment with other methods, but be prepared for failure and frustration.

I agree w/ tock

But if you're desperate to grow or sterilyze, then people shouldn't forget that those are Options..But options that you should

I dunno sometimes I find it quite annoying, i guess cuz mine is so big heheheh.

i would never do any bulk in oven or microwave tho, these were basically just experiments for pc-less ppl to take note of.

Its always worth the try.Without experimenting we would be no were.
Better luck next time.

Lol

It's not that it doesn't work, it's just that ir's not as "reliable" as other methods (boiling ect)

Enough contams with a pressure cooker possibly. At least with a pc you are more then likely sure no contams, then you can worry about other stuff. Unless you have the ultimate sterile environment cleaned by professional lab people lol.
