Mycelium
    #3946859 - 03/21/05 12:40 AM (12 years, 8 months ago)

In Stamets book he seems to think the it's best to start Mycelium in agar, then cut it up and colonize jars of substrate with little pieces of mycelium. Are my chances for succes better if I do that, or just try to colonize substrate right away?

i wouldnt say your chances are better. The reason he does this is to isolate a fast growing mycelium for faster colonization times and better yeilds. its alot of work. If you have a print you shold make a spore syringe. It is super easy and has a pretty high sucess rate. You will get great growth. Once you get good at this you can put some to agar then transfer that to a liquid culture.

So... It's better for success rates to turn a print into a syringe? I'm trying to get Cyanescens or Azurescens, but the only spores I can find of said varieties are on prints.

well first off lets make sure you live in an area that they will grow in. Now get your prints from www.sporeworks.com I know that you can take spores from these prints and make a sterile syringe with very little effort. if you do try agar there is no need to isolate. Spores to agar, then make a mycel syringe from that and knock up a Karo/dextrose/honey jar with that. You want maximum genetic variation for your outdoor bed because you don't want months of work to go down the drain when it doesn't fruit.
Anyways get your print from spore works, soak alder saw dust from a BBQ store for an hour let it drain, add wheat bran at a 4:1 wood:bran ratio, PC for 30-60 Min's and then knock up with the spore syringe that you made. It WILL work, you will be fine. Make 12 jars with that spore syringe and you will have plenty of genetic diversity and you will have plenty of spawn for an outdoor bed

in this pic the person makes a not to insure that this is azure. it would suck to accidently mix up another species as azure and build a big ass outdoor bed only to get nothing

I'm in zone 5 (S. Wisconsin) Would an outdoor bed be a waste of time?

an outdoor bed would probably be okay but it would have to be covered in the winter. You might want to try cyanofibs, they seem to be more adaptive to different climates

By Cyanofibs you mean cyanofibrillosa right? Would SporeWerks be a good source for them too?

for sure, they have them
Sporeworks is your best bet for these types of spore prints

Tahoe, thanks a lot for your help. I have tried cultivating before, and only ended up w/ green slime in jars. I am thinking of doing it outside, because it's a lot safer in a legal sense. I guess @ the first of the month I'll order a cyanofibrillosa print from Spore Works. I think I'll try for a cyanescens print too, the potency intrigues me . But you say that my mycellium could be perennial if I cover it, that excites me. do you think mulch would work?

You can buy a copy of Stamets & Chilton at Amazon (used) for $22.00 plus shipping. It's not current with all the great stuff I read about going on here, but it does put a good foundation under you.
