But look at it. It's all boxed up and can't get away. I've never seen it go this long without forming satellite colonies. The reishi is definitely holding its own.

Hello, all!

I know I'm a Newbie here but

A) I had to bust my cherry
B) I have some experience with moulds in culture
C) I'm interested in the way Basidiomycetes behave on agar, so

What colour is the reverse of the "green" colony? The obverse looks Olive/~Brown to me, suggesting Cladosporium, not Penicillium. If the reverse is dark this is most certainly the case and you may or may not have a viable culture. Some Clado. spp. will restrict other fungi while not necessarily inhibiting their ability to fruit.

Hope this is a helpful first post.

it's penicillium, alright.
though my exp. with it has been blueish, though i have never watched it culture on agar.

i can't wait to see what happens next episode.

It may possibly be aspergillus or even Cladosporium, but I'm filming the action for a time lapse video and I don't want to disturb the placement of the dish to get a microscopic identification on the contaminant. It was a green color earlier, but has since faded to what you see. If you'll look at some of the earlier pictures, the blue/green color is a bit more distinct.

Ahhhh, true true...

In the first couple of pix the colony is like a deep aquamarine...? I've seen both Asp. and Pen. express this colour. Sometimes Clado will also look this way VERY early and depending upon media and lighting (I'm used to MEA and halogen/fluorescents).

It looks as if the exercise is more a matter of entertainment than anything else so it doesn't really matter, and you can't really ID a mould based on colony morphology alone... You're right to not disturb the plate and invite satellite colonies.

If you're interested in naming the intruder at the end of the clash, PM me and I'll be happy to look at her under my 'scope.

Something I have not seen so early has happened. The agar dried out after only two weeks and was starting to pull away from the edges of the petri dish like the dog eared pages on a well-worn book. The reishi and the contaminant are both fighting to the death here. I injected some sterile distilled water onto and below the agar to re-hydrate it. More pictures coming.

I doubt it. I use halogen lighting, but it's on a cycle timer to only come on for 30 seconds every twenty minutes when the vid cap is being taken. I think it was the battle. When I get back to the lab later I'll get a pic up. Most of the sterile water has been absorbed by the agar.

Ok, here's what happened. The dish, unwrapped to keep it from fogging up earlier, dried out. Look at the edges peeling up at the six and twelve o'clock positions.


After adding water to the agar, it seems to have re-hydrated. Hopefully, growth will resume soon. (The first and last time I'll ever root for a contaminant)

The reishi has succeeded in containing and isolating the contamination by absorbing all the available water in the agar. Look at the above picture before added the water. The edges were curled up from being too dry. I haven't seen that before from dishes that were less than several months old. I will consider when growth of both organisms stopped and the dish became totally dry as the end of the experiment. I know that dry reishi mycelium will start growing again after at least two years. I don't know how long the contaminant would remain viable.
