Crap colonization...

I've had two successful batches of PF style BRF jars, and trying to get my third going. This batch was inoculated using a syringe that I made from a Cambodian print from a friend. The syringe was allowed to sit for about 20 hours before I used it, so the spores in it should have had time to rehydrate. The 8 substrate jars I made were allowed to cool overnight before inoculating. About 1mL of spore solution was injected into each jar. It's now been 12 days, and only two of the jars have germinated, and only in one spot each. 6 of my 8 jars have absolutely nothing going on, not even contamination. The only difference between this batch and the two previous ones (which were both very successful) is that I used a homemade syringe made from a homemade print. If there are no contamination problems, then I don't see how there is so little germination. Any comments on this?

It's hard to say man. There could be a problem with a bacteria causing no germination. If your syringe was rich in spores, this is could be the problem. But there are so many other factors involved that could play a part too.

Did you hydrate your spores for at least 24 hours?

unfortunately i don't have an answer for you. but i have a similar situation. i just made my first syringes(from a good source)and i am so paranoid that something is gonna go wrong(contam).guess I'll just wait and see. good luck!

Odd problem. I'm betting you didnt shake the syringe before inoculating? That helps get even growth alot. Perhaps you messed up the water content in the jars a bit? Little to wet on this batch or something? Takes spores that long to germinate if you make them extra wet, or dry..sometimes.
And they may not have been rich in spores..which, to whomever asked just means the person may have just used a small amount of spores in the syringe.
The print also could have been older, affecting growth.
And do you shake the jars when you inoculate them? This is grain right? I forget if were talking grain or PF, but if you shake the jars after you squirt in the spores, I'd suggest not doing that and shaking the syringe instead. I've found I get quicker starts when I inject if I dont shake the jar right after.

These were BRF/Verm jars, so no shaking. The spore print was fairly fresh, about a month old. The water content was exactly the same as the last two batches. The syringe was made from half of a spore print, so it had a ton of spores in it. The only thing I can think of is the spores didn't get long enough to rehydrate, but 20 hours seemed like enough. It's pretty irritating, because my first batch of jars is on its third flush now, and it looks I may not have enough reinforcements to replace them after they're done.

sorry to get a little off topic, but is it common to use 1 print for 1 syringe? that's how i do.. am i wasting spores? or am i speeding up my colonizing time? my syringes are purple and you can barely see through them....

The print could have gotten hot sometime killing (almost) all the spores.
