    #3888919 - 03/08/05 08:22 PM (12 years, 3 months ago)

last attempts to isolate from gill fragments left me with a weird mold/fungi that colonised like quick silver and formed microsclerotia by the houndreds. But it wouldnt take to straw. Dont know if it was fungus or a mold to tell you the truth eather. So i want to get fruits this year and if any one can help me with my endevours it be much aprecited and well compensated.
Thanks for taking a looksy
peace

I will throw down a share on a culture from Penn State Labs, if it comes down to it.

Shirl said she could possibly throw down on such a venture.

http://www.dsmz.de/strains/no006947.htm

Penn State only has wild volvariella cultures. That is a cloned off a commercially grown mushroom.

They don't have siniensis

http://www.dsmz.de/species/fungi.htm

On edit: Its too cold here for me to receive this culture.

If the price is right and we have enough people, I'd be in to thrown down on a share of a Volvaria culture as well...

But of course, I'd have to wait for temperatures to warm up before shipment could happen...

If anyone would like to attempt an isolation on agar, I can send him some dried gills.
You must be versed in the use of agar. PM me.

Maybe if we get some benlate that would be a realistic possibility?

I think I can find dried paddy straws locally.

Okay, I can go to Penn State to get the culture.

Hopefully this can happen around the week of May 22. My friend from Maine will be visiting me. I can meet him at Penn State and we can check out the lab. Then we are going to go hunt morels, bluefoots and whatever else we can find. After which we will be heading back to Indy or further north by caravan to find more mushrooms.

Is it true that Volvaria cannot be kept in the fridge because thought they are thermophylic they can't take the cold? This would make preserving a strain difficult because of the necessity of constant transfers. Let me know what y'all think. Also, I would also be willing to throw some money down on the strain.

Negative. My first attempt resulted in some very nice bacterial cultures, but nothing fungal. I sterilized my loop, cooled it in agar, and then swiped a very small sample of the powdery stuff that was clinging to the inside of the bag. I then streaked this out on a few MEA plates and incubated at about 80F.

Perhaps some peroxidated agar would be useful for my next attempt?
