extracting liquid mycelium into a syringe

i have a honey jar that i inoculated with B+ 2 weeks ago in my incubator. it seems to have colonized quite nicely, except that the white fluffy cloud is sunk at the bottom of the quart jar. 1) is this normal? that is, is it normal for a liquid culture to sink, or should it be floating?

also, when i want to extract the culture into a syringe, 2) how do i get just the mycelium and not all the water? 3) should i construct a glove box for the transfer of liquid mycelium to a syringe , or would it be okay to do in an open yet sterilized (as much as possible) environment? i suppose if i'm going to get serious about this i need to build a glovebox in the first place. i'm just really cheap, and not too interested in spending money i don't have on a "hobby," and any hobby at that. thanks dudes.

2 you dont, some of that will be the carrier for the mycelium, you can stand it on
the plunger and wait for it to settle a bit and evacuate some of the solution,
leaving mycelium behind.

If you shake/ swirl your jar every other day or so that really helps break up the myc and spread it out so it's not hard to suck up.

Oh yeah! A syringe with a lot of tissue from mycelium is a lot more effective and faster of a colonizer than a clearer syringe any day of the week.

next time you make a honey or karo jar up :

put a mable in the jar...this will help you break up the mycelium when you are ready to make syringes.

make inoculation ports in your lid...put a rubber(injection port) or some silicone over the hole in your lid.
then you won't have to open the jar in open air.

I put another hole in the lid for air exchange...I cover this hole with a small round bandaid.  :smirk:

Example:


You can see the marble inside the jar.
If you look real hard you can see a dab of silicone ontop of the jar lid.

the thing is, i used a 1 quart jar filled about 2/3 of the way up, not a 1/2 pint jar. so there's a considerable amount of water that i'll have to be sucking up. sooo...what if i had the liquid culture jar and 2 sterilized syringes and some sterilized jars of grain in perhaps a glovebox. as my grain (rye) has always come out a little dry, what if i injected a whole syringe or two into, say, about 7 jars--as many as my pc can hold. i'm sure i have enough water in my liquid culture jar for 7 jars. but the issue is this: would this work?
