colonization problems and possible contamination
    #3774803 - 02/13/05 04:11 PM (19 years, 2 months ago)

AFOAF recently purchased some spores from hawkseye, and even more recently innococulated 2 (6 jar) batches of jars (totaling for 12 inoculated jars)
he used the PFTek for Simple Minds guide, and incubated them in a TiT incubator at around 85 degrees F.
the first 6 jars he inoculated showed no signs of mycellium growth after a week, and the clumps of BRF in the jars slowly discolored to a crimson, almost blood-red color that slowly spread and became more apparent throughout the next two weeks of attempted colonization.

the first batch's holes were completely covered by tinfoil, and the second batch's are still all covered with surgical tape.
AFOAF is currently experiencing no signs of mycellium growth after three days in the second batch (not totally unexpected), and they've been incubated (along with the old jars, which were completely covered with tinfoil)
at 83 degrees F.

on a separate note, AFOAF used very clean techniques in innoculating, including the Oven Tek (at around 200 degrees, set on the outside of a pulled out rack and inoculated), gloves, and masks. not to mention soap, and disinfectants.

so, my two main questions are:
1. why did AFOAF experience no mycellium growth after 2 weeks of incubation in the first batch?
2. what is this red discoloration spreading (in areas of the jars where AFOAF injected the spores) in the jars? it doesn't have a mold growth, and does certainly not look like fungal growth, it's just a discoloration.

any comments would be greatly appreciated, since this is a troubling first attempt, and hopefully the money AFOAF spent on this project will not go to waste.

The world is like a ride in an amusement park. And when you choose to go on it, you think it's real because that's how powerful our minds are. And the ride goes up and down and round and round. It has thrills and chills and it's very brightly coloured and it's very loud and it's fun, for a while. Some people have been on the ride for a long time and they begin to question, is this real, or is this just a ride? And other people have remembered, and they come back to us, they say, "Hey - don't worry, don't be afraid, ever, because, this is just a ride... And we... kill those people.
