 Big-time contam problem.Pleeeez help

I just innoculated 9 jars and only got 2 without any contams,, i use an alcohol flame but can`t seem to get the needle red-hot all the way. My question is would butane flame work ? ..like from a pencil torch. not a cigarette lighter.

umm.. my needle never gets red hot, and I have not had contam problems like that.. you sure the needle is the source for the contams? How did you sterilize the substrate? and did you get the spore syringe from a reliable source?

Sterilized for 1 full hour at 15 lbs starting the timer after the weight starts hissing....seems the contams are exactly at the point of entry of the needle directly below the taped holes.This is why i asked about getting the needle hotter.. My resources for syringes are PF and sporeworks. never had this much problem with contams before

Your contam didn't neccissarily come from the needle. You don't have to get it red hot either, just a quick 3 second flame will do. Since you did flame it good, I doubt the syringe is the problem. Unless you only flamed the needle once? Flame it right before each jar, better yet, each hole.
Did you pressure cook the jars? Or boil them real good?
Make sure you have a real clean environment. Read some "sterile tips" at The Hawks Eye

I did heat before each hole..... would cooling the needle in lysol maybe help?

verm barrier bro yup....... wipe the inner edge of jar clean then add dry verm . about a half inch.... really puzzledn why i get green mold right at the needle point of entry

That's a tough call. Is there anything you've changed since previous, successful crops? I tend to doubt your needle flaming technique as being the problem. I sterilize my syringes simply by setting my syringe in a beaker partially filled with alchohol.

I do the same thing and just flame it on first use and last before putting it away. then put in alcohol between holes/jars.

just to figure out where the contamination is coming from, you could do some negative controls. . .with PF jars it's easy since you can make them so easily.
Sterilize everything as normal.
Keep one jar aside, don't inject anything, but incubate with the others.
Take another jar and stick the needle in each hole, but don't inject anything [dry inoc.] Incubate as usual.
Inject the others from the same pressure cooker batch normally, and you should have an idea of what's going on. If you REALLY want to go all the way, do two or three of each negative control. If they don't contaminate, you can always use them later.
I'd also agree that it's probably not the needle itself giving you problems.

I'm completely new here, and haven't had too much experience yet, just a LOT of reading and a few jars currently colonizing. In fact, this's my first post, so.. don't take anything I say too heavily..
Your situation is pretty confusing.. seems like you're doing everything right, but there're a few things that come to mind if you haven't found the answer.. Have you tried cleaning the whole syringe really well? When you put the tape on, are your hands clean, or do you at least not touch the part exposed to the inside of the jar? Are the holes clean-cut so nothing can hide in the edges? How long do you keep the tape off when you Innoculate? Another thing, though kinda stupid sounding.. do you ever remember possibly sucking air into the syringe or anything that might get contamination stuck in the needle?
If you've checked all of those off, I really can't suggest anything other than an innoculating hood, a LOT of lysol, and wiping the needle with alcohol too..
I know most of that is just logic, but I hope some part of that was helpful

i'll try to help as much as i can.. you said you innoculated nine jars and got two w/out contam.. i think i know why. nine plus two is eleven.. i have a little brother who is eleven... when he doesnt do what he is supposed to .. we keep him in his room till he is better.. so i would suggest you put them in the corner or something till they do what they are supposed to .. if that doesnt work try to put pepper on their tongues. glad i could help.

just a idea ,but when you are done pcing your jars are they on tight with the tape on them .because if that is the case when you go to put the neddle in there is a vacum in the jar might be sucking in the contam that way.you never know i just thought i would try to help,hope you get it worked out :-)

If you're still unsure about what procedure went wrong when innoculating then try using a piece of cotton with alcohol, put the cotton piece above the hole then innoculate, so the needle goes by the cotton+alcohol before entering the jar.

Thanks for all the informative replies... I do remember the syringe getting stuck and drawing it back a hair to unstick it...... wonder if that could be the problem?.. anyways...I just got 2 new syringes from Sporeworks and going to knock some jars up tonight. I think I`ll try the inserting the needle through a piece of cotton soaked in alcohol before entry. that sounds like a good idea and i`m sure it couldn`t hurt. Thanks again for all the response

Yep.. I bet that's what did it..
I guess the reason why this wasn't all that obvious is because the newer cultivators have a HUGE fear of contams instilled in them from all that they hear about 'em.. When I got my syringe I was afraid to even TOUCH it, let alone use any moveable parts.. lol
