How do i achieve proper water content, while fractionally sterillizeing grain?
    #3689832 - 01/27/05 07:44 PM (11 years, 10 months ago)

Im trying fractional sterillization for the first time, becuse i no longer have access to a p.c., im useing bird feed as substrate and i was wondering, should i steep the birdseed before i start the fractionall sterillization?P and T for your responses

I don't know what fractional steri. is...maybe boiling?
If that's the case then I think it would be next to impossible to do that and be able to call the substrate 100% sterilized.
As I understand it, you'd be lucky to kill half of the endospores, or even a fraction (no pun intended) with that kind of heat.
If you don't have a P.cooker at your disposal, then you'd be better off doing the PFtek which uses brown rice flour.
Maybe others have tried this method with some degree of success, but certainly not constant.

I think it would take alot of trial and error on your part and if you switched grain type you'd have to start all over. I F_S'ed some WBS and it turned out very mushy. I was trying to sterilize with out a PC cause the one I had sucked!

Fractional sterilization involves boiling the WBS three times, one time a day for three days. Its supposed to work sometimes.

Water content: The overnight soaking of the grain then the 20-30 minute simmer takes care of the correct water content for you.

The problem comes in the three boils(water content) unless you watched closely and boiled at as low a temp as possible. I don't know JMO, it must work or people wouldn't make a tech 4 it, but with grains there is too much to mess up IMO-

You can do it either by soaking/simmering the grain first, or by adding the measured amounts of water and grain to the jars directly. Should you notice that the grain is too dry after the first boiling and shaking, you can add some more water.

thanks anno, thats more of the kind of answer i was looking for. Do you think that fractional sterilization actually works? Because theoretically its all there. you know the whole germintation of threatening endospores, and then killing them off with later half hour boils.

I tested it out a couple of times with rye, it worked for me.
But it takes long, so if possible, try to get a pressure cooker.

Anno would you recommend letting the jars used in fractional sterilization sit for about a week or so before inocc. to see if any contams grow? Or would that be worse?

give them 4-5 days of rest to check for contams, if you add a little gypsum it
will help to stabilize the water content of the grain and prevent bacterial contams

It doesnt work like that. The 3 boils dont go in place of simmering..your boiling the jars filled with substrate 3 times..in place of pressure cooking. The water content should not change from before your first boil to after your third. First get the water content where it needs to be by soaking and/or simmering...then load..then sterilize.

Has anybody had success with this yet besides Anno? If your lid is loose water may be lost on each boil. My idea was to fill bags or large jars with grain and microwave them. 1ml of water weighs approx 1g so you can weigh your jars and grain and add excess water. Then boil it down over a few days until it weighs exactly what it should with your correct water content. This way you can handle large bags without the need for a large PC. I had success in sterilising rye grass seed, rice and wheat, got some bags started but lost them to contamination. This could have been contaminated spores since I made the syringes myself and was not overly careful.

until then, good luck with your results on the fract. sterilization.
