hey all i am in need of some help(ONCE AGAIN). So i inoculated 3 mycobags three weeks ago they are in the incubator at 86 degrees now and i have 0 myc growth? i put in 3 cc,s per bag and after 2 weeks i put in a little more. I have no idea what I'm doing wrong ? do the bags have to be fully unraveled so the air patch isn't folded over? Because when there in the incubator there isn't much room so the air patch kinda gets folded over in the bag. This is the only thing i think could be wrong. well sorry for the ramble but i really need help........thanks ...B

hmz the airpatch is there for a reason so yes let it do its work But even without it you should have seen some growth after 3 weeks....
I dont know whats wrong! Tell your procedure please in detail...

Humm... 0 myc growth after 3 weeks... I think in a dead spores by sterilization mistakes.

ok i ordered 3 different strains to try em out i got GT, hawaiians , and b+,i ordered a grain substrate mycobag (3). I tried to make everything really clean like the tek's say i first flame sterilized then wiped cool with an alcohol swab, i also wiped the inoculation point of the mycobag with alchohol ans then put them in the inncubator. At first i had a problem with my inncubator and things got to hot . I thought i may have either gotten things way to hot, or maybe i didn't wait long enough for the syringe to cool before i inoculated, so i then inoculated again. they are now in the inncubator and at 86 degrees (TIT). i think its time to put the grain in a cereal bowl and add some milk

could it be a bad spore syringe? i don't even get mold growth? How could i have killed the spores in the syringe. I got the syringes from one of the sponsors online , i guess they could be crap?

Ok too hot (somewhere 120+)kills spores 100%. Can imagine viability decreasing already with high 90-s and upp. Also depending on how long they were exposed to high temps.
Those bags come sterile dont they? Did you shake the syringes before inoc? Dont know about the weather in your region and which route the syringes took. But can imagine that along that way cold and heat CAN have played a part as well...

well i live out in the cold N.E and they were definitely cold during shipping . and i shook them up real good before inoculation. my incubator was hot but i dint think i got the bags up to 100 degrees.

You have to remember that the subtrate will produce heat also. If the chamber was at 86 the contents of the bag was a bit higher. Too hot is worse then too cold.

OK say i did ruin the bag at first getting the temp up to high, but i then inoculated again after i fixed the heat problem . wouldn't this be like starting over? its been almost 3 weeks since SECOND inoculation.

umm it has condensation on some of the bag, its not really wet but definitely like morning dew

hmmm. it may be that it is too wet, or that your syringe is no good. when you inoculated, did you inject into the center of the substrate or on the side next to the plastic?

To wet or to dry, doesnt matter....SOMETHING shouldhave germinated by now if all you've said is correct and spores are viable. Did you only try one syringe? Make sure you attempt all 3. Did you buy from a sponsor? If not, that could be your problem. If so, it has been really friggen cold up here in NE recently...cold weather could have done it...I've seen a number of people with frozen syringes and no problem...weird it would kill all three.
You definatly should keep it unwrapped as well...let the filter patch do its job...I still dont think this is enough to STOP germination.

i got three syringes form the vendor and i used all three .could the cold weather have killed my spores? at this point i would be happy to se mold grow
