blending agar for liquid inoculant
    #3676152 - 01/25/05 05:46 AM (12 years, 4 months ago)

ey!

how much of a standard petri's content would you blend up in sterile water
in order to do a couple of syringes, say two or three?

oh and jeah, say you fix such a syringe and slam it to the fridge with
some air left inside the syringe, could that baby be stored like for
a month or would that be a bad idea?

A wedge.

And, don't slam the syringe against the fridge, it might break

I've heard of people having success with syringes made with living tissue for up to a couple months if kept in the fridge. I'd say a month is safe, but you want to be sure that they won't run out of nutes. Maybe make some honeywater and get that sterile?

so you'd recommend blending them in a VERY light honeywater mix if i wanted
to store them a bit.

wouldnt this addition of nutes, which obvously wouldnt be present in
the just belnded wedge-water, also get them to produce CO2 inside the syringe? --> new problem..

Yeah...and that is one reason you should NOT have any nutes in it when you store. Things put into storage should not be growing and therefore should not have any nutes. Cultures go into storage once all nutes have been used up and it stops growing.

You can use a few drops of h202 and it will last 2-6 months (maybe more but thats pushing it) stored in the fridge.

the needle has a hole  Maybe contam risk with using that. I think myc syringe can be stored in fridge for a month or 2. Just like one can do with colonized jars or agarplates. Dont know if the being in te water shortens the period but I guess 1 month must be very fine.

They dont need nutes then cause they hybernate c.q. dont grow, so dont need "fuel".
