Agar inoculation?

I have read all of the tek's on making agar plates, then the grain, then substrate, but all of them start with the agar being inoculated by a piece of tissue from a already fruited culture. My question is whether or not you can inoculate an agar plate with a spore syringe? It seems to make sense to me just wanted to make sure and see if anyone else has done it and has a word to say,

Yes, you can. But not if you use peroxide agar.

oh yah, about how much of the spore syringe should i put on each plate?

A drop or 2, and then tilt the dish so the drop spreads over the agar surface.

CAnt add anything. Just wanna ,ention that I tried Doc's34 alt. agar tek and it workd wonderfull.
The rhizomyc I transferred to liquid medium and it grew VERY fast! :thumbup:

I tried a few different methods for innoc of agar via syringe, and I noticed:

Injecting INTO agar: 4 weeks and only some very small spots of mold have developed in random areas...can't even tell if it contam it is so small.

Injecting ONTO agar: after only a week healthy mold showed on 4 of the 8 plates. All plates had condensation on lids, and after shaking the condensation back onto the agar surface, the other 4 kicked in.

Conclusion;

Injecting into the agar needs some more testing and controlled environment

Injecting ONTO the agar is a sure way to get mold, however it is possible that progress could be slowed due to condensing spore liquid....doesn't make sense, but that's what I found.

I have used spores on agar several times to start new strains of Cubensis.

John Allen (MJ) my good friend, has brought back new strains of mushrooms from around the world and has given me some to start...I am truely blessed to know him.
I always start with his spore print and put them on agar...then sector them out til I get very nice rhizomorphic plate...then transfer more til I'm happy with the results.
Then I grow them out on cakes and casing to see the results.

The Allen strain, Elephant Gate strain, Burma strain and Malabar strain were all started this way.

I have a few new strains that haven't been grown out yet...soon...very soon.

ofcourse not IN the agar, just dro psome spores on the surface of it.

If I am correct a clone grows better if embedded In the agar, but also that worked very fine ON the substrate...
