as no one has replied in the cult forum, i thought maybe you veterans could enlighten me
1)if you inoculate with fresh tissue, is this the same as cloning ?
2)is cloning used to create an even pinset/flushes
if i am confused about this, maybe someone could tell me the applications of cloning in mycology
my apologies if this was posted in the incorrect forum

jars or
do you referr to tissue on agar?

i guess your talking tissue out of the mushroom, this would basically be
a clone yes, since the mycelium growing on your grains will be the one
that was in your mushroom - same dna.

cloning is used to, well, clone a mushroom and its characteristics.
such as shape, look, size, taste or whatever.

you could get a way more even pinset with clones yeah.

can you inoculate wbs with a chunk of a mushroom instead of grain to grain or knocking it up?

Yes and yes. You can clone to both seed and agar. You clone to get good pinset and disreable charateristics. So pic one of the first shrooms to appear and mature into a nice specimen, clone it. It will be an early pinner.

Illahee, you can clone that way. You need to but a shroom in half and take some tissue from the cap or the base of the stem. i just did this with some portabellas. Tissue to grain can contam easily, tissue to agar is better

but people dont realise that your only cloning a type specimen with a variety of possible phenotypes - a variety of possible forms that it may take on cultivation. You can clone the best looking pinset on earth and it may recultivate and express a different, crapper phenotype and you have wasted your time.
therefore, cloning mushrooms isnt really cloning in the sense that we would understand of the concept.

Can you clone by taking a piece out of the base of a shroom and throwing it in to a karo jar? Will this create a liquid culture of the clone, or not work?

yes you can clone that way. Just make sure you keep that peice sterile. i took a pin from a petri and put it in karo

Dunking the piece in water/h202 mix prior to adding to karo should help keep it sterile well enough given a sterile environment, yes?

i am not a big fan of using h202 as a sterilizer. Cook up your karo with a tyvek top. Put you karo and a mushroom in a glove box. Tear the mushroom in half, heat up something and poke a . size hole in the tyvek maybe a little bigger. Take a small piece of flesh from the cent of the shroom and push it through the hole. Make sure it hits the karo. Tape off the the hole and give it a few days to swirl it. You should be fine

I think i would avoid trying to nock up a PF style jar with tissue. Your best bet would be a Karo liquid culture or an Agar petri dish. I take a pair of tweezers that i use for mycological research ONLY and pc them for 25 minutes @15psi wrapped up in aluminum foil. Clean your gloove box with soap and water with a clean sponge, just for that use. I have certain sponges that i use for particular things like, cleaning gloove box, tools, wiping down jars and dishes, and one for wiping my arms and hands. Put your wiped down jar of Karo in your box along with the foil wrapped tweezers. I like to wipe the foil down too with a sponge. Now pop the cap off your clone of choice and give the stem a wipe down with a sponge of soapy water too. You want to be able too squeeze the hell out of the sponge with no soapy water comming out. Set it in your gloove box. Now it's time to clean your hands and fore arms. Sound like a pain in the ass doesn't it! But it really isn't once you do it a couple times. Being clean is key for work like this. Don't cut any corners. Now loosen the Karo jar lid to the point you can take it off, but don't. This will make it easier to put the tissue in the Karo. Now unwrap the pair of tweezers and leave them in it unwrapped but to were you can just pick them up out of the foil. Now crack the stem in half, DON'T cut it in half. Tear it in half. Now pick up your tweezers and pinch out some tissue out of the MIDDLE of the stem. It's nice and clean in the middle of the stem. Now lift up the Karo lid and drop it in. If you've loosened the lid before hand, you don't have to fiddle with unloosing the lid with tweezers in your hand. You see what i mean? Tighten the lid up on the Karo jar and relax! It's tedious.You'll start to see growth of the tissue within a couple days. Allow it to grow for a week and then start swirling it. Putting pieces of glass in your Karo solution before pressure cooking works great for helping seperate or break up the growth when gently swirling it. Hope this helps. Good luck! It's not as bad as it sounds. Go through the process in your mind and plan ahead. It's no poblem after a couple trips.
