LIQUID CULTURE FILTER

Optimal Liquid Culture REQUIRES GAS EXCHANGE:
THE PROBLEM BEING, any filter material gets wet when you agitate the jar. Wet filters wick in contamination & the culture goes to hell.
A simple - inexpensive way to circumvent that problem:
Requires plastic lid that will withstand PC'ing, drill & bits, polyfil, surgical tubing, stainless steel screw & hemostat (or whatever to hold tubing shut). The pictures tell the story.

good idea! i always thought polyfill worked poorly with liquid culture, and this is much cheaper than filter discs. 5 for you if i haven't already rated you. also, a rubber band might work to hold the straw shut, just an idea

It's rubber SURGICAL TUBING, which will withstand PC'ing. A straw will not. Polyfil isnt the best fiter material. But, years of its use in the PF tek, show it works.
It works better in this aplication, as the surgical tubing HOLDS it TIGHTLY in place.

You can also do it like this: Here is why. A syringe fits snug into the rubber tubing & you don't spill a drop.

I must of had a brain fart . Was thinking this http://www.fungifun.org/polyfill/ (polyfil tek) & instead said "PF" tek. Glad you are on your toes. I'm to tired to be on mine, right now.

well, if you take a jar as the one you pictured above and equip it with a ghetto self healing injection spot and a nice tyvek filter you can do just the same without all the funky lab tubing and whatever sooo lets take agars nice but complicated liquid culture method and ghetto-ohm-simplify it what you need: small jar with metal lid screwdiver hammer tyvek silicone glass-sheds liquid culture media sporesyringe nail allright, you punch 2 holes in te lid, a large one with the screwdriver and a small one with the nail. use the hammer do flatten the metal pieces standing away after punching the holes. no will teh jar with the culture media and throw in some glass sheds. cover the jar with tyvek and screw one lid loosely to check out where the big hole will be. now, with lid just like 80% on, smear a LOT of silicone over the big hole and make sure there is some under the lid aswell, you want it to touch the tvek beneath. screw on tightly, make a nice round silicone roll and let sit for a day. now autoklave it, let cool, wipe SELF-HEALING silicone injection site with ipa, inject spores and let colonise. move it around gently every few days, not shaking it, that will distribute the tissue very nice since the sheds will do their part cutting up the tissue real good. no need to rougly shake it and wet the tyvek, just be calm like with a lady sooo after some days or a week or whatever you like, wipe SELF-HEALING spot again and draw up sringe. the small hole served as an air exchange hole during colonistion and now filteres the air entering the jar as u draw up the syringe and create negative pressure in the jar. hooray for ohmteks rofl oh jeah heres a pic peace ohm

I use these to make Liquid inoculate.
It's PDA .I slant it almost flat, grow it over with mycelium.When it's covered I break it up with a sterile(torched)stiff wire and fill it up with sugar solution water and sterile water...shake like crazy for 3 days. I make these well in advance and seal them with saran strips and tape..before I wrap the ends I dip it in isopropol.
the shaker thing is a 40 rpm 120 volt ac gear motor from princess auto.I clip the tube to it and let the air bubble roll around for 3 days...about a third of the agar dissolves into the water and it makes a liquid soup food full of mycelium that takes over creepy fast.
one big tube makes about 10 CC of liquid food myc suspension. yeah it's a one shot thing but if I need I can always just tape a couple more test tubes to the rotator.and I don't have to worry about cross contaminants.
after I drain it,I put it back in the incubator and let it grow over again then inject more sugar water into it.another 3 days on the spinner and it's good to go again. just my 2 cents

i dont get it why everybody does so complicated teks just to get liquid media done.
i mean agar / peter's teks sure work fine, but the way i do it sure is a lotta easier and prolly gets me just the very same results as the other two teks..
i mean 'lab grade' tubing and even electrican 120vol gear motors for shaking - lol dudes, this is mush cult, not rocket science peace ohm

I think alot of the ideas agar has are marketable. It would be one shroom kit nobody could make fun of. I think the ideas are cool even though agar doesnt answer my questions.
Is all rubber surgical tubing autoclavable or do I need to find a certain kind of rubber? AGAR???

I could not say ALL surgical tubing is autoclavable. But most rubber tubing is, & almost ALL medical type grade rubber tubing is.
BTW........agar is working on a KIT.

Nothing complicated, no 'lab grade' tubing required. Fast, simple & easy to build. Took me about 70 seconds. Will work with pint, quart & gallon jars.

Is dry dextrose/ligth-dry-malt combo. Weak solution 1 teaspoon each to quart distilled water. Hot solution = double that. Super hot = triple that.
