Grain problems, jars not colonizing
    #3596592 - 01/08/05 12:07 AM (19 years, 1 month ago)

I inoculated 20 quart sized grain jars on 12/18, using Mag's tek.

As of 12/27 a lot of the jars had shown no signs of mycelium.

So I took some caps from my brf cakes, and did some prints to make new syringes. This was based on the assumption that my jars didn't actually get enough spores originally.

My method for making the syringes was to take a nice size cap, place it in a sterile half pint jar, seal the lid, and wait. The cap made a nice dark print in the jar. I removed the cap, let the print dry for a couple of days.

Then I boiled some water, sucked it into a syringe a couple of times, then allowed a full syringe time to cool. Once cooled, I removed a bandaid over a hole in the half pint print jar, and squirted the water in. I did this a few times. Shook the jar vigorously to spread the spores out.

Then I sucked the solution back into the syringe. Many clusters of spores were visible inside.

On 12/30 I re-inoculated several jars with these syringes, making sure I shook the syringes well. Each jar was inoc'ed with 2.5 cc's.

As of today, my jars STILL appear to not be germinating.

I am pretty certain that all my procedures have been as sterile as possible.

Anyone have any ideas what the problem is?

I have also tried to G2G a few jars, but that was only 3 days ago, so I cannot really be conclusive on those results yet.

Thanks in advance for any input.

I forgot to add that my incubator is where it should be. Its constantly 82 - 85 degrees and completely dark inside.

You used rye right? IME, lack of germination has been due to a high water content. How do they appear on that front?

G2G rocks!

Also make sure your giving the grain enough time to cool down after the pc, I never do anything before 6 hours is up. Which reminds me, make sure you are shaking the jars as soon as the pressure is down in the PC. They will be very hot so you'll need pot holders, but make sure to shake well to disperse the water evenly. Also make sure to shake well after you inject the spores.

My guess is that the water content of the grain was not correct.

I'm in the same boat and I have to lean toward a little to wet. In most cases if they are a little dry they will still show good signs and then stall at a later time. When wet they can take forever to start if at all.

Some things to look for.

To wet.

Was grain soaked long enough (24hrs).

PC long enough. (1hr at the very least at 15psi) You can kill the bacteria on the grain but not in it if you don't pc long enough.

Feel free

How can you prevent the water content from being to high. I simmered them for the amount of time that Mag's tek said.
