(TLG) UTRASONICLLY TREATED SPORES

UTRASONICLLY TREATED SPORES & SPOREBAGS:
Agar long ago received his first successful spore batch from TLG. Over the years, he received many differing strains from TLG, without ever a problem in orders.
TLG gets fecal matter thrown at him from another vendor - because TLG is introducing into his inventory NEXT GENERATION, I., E., Ultrasonically treated spore syringes. Which is said to disperse spores into a solution incredibly well.
Well, agar saw so much fecal matter thrown at TLG, he decided to have his foaf give it a test. Just to see for himself. Agar?s foaf makes PRINTS on autoclave bag material (SporeBags). He does so for several reasons.
1. It is easy. 2. Autoclave bag material is inexpensive. 2. Autoclave bag material can be SEALED with an impulse sealer. Net result is a print, sealed in a sterile waterproof material, that is impervious to contaminates.
To test TLG?s UTRASONIC DISPERSAL method. Agars foaf used a piece of autoclave bag material measuring 6 inches wide & a 12 inches long, then set 2 big fat healthy EQ caps on it & made a double print - which he allowed to dry 3 days.
He then folded the bag in half (prints facing each other) & impulse sealed the bag. He then injected 40ml of sterile water into the bag (on a corner), then resealed the bag (impulse sealer). He then dropped the bag into an a water filled ultrasonic cleaner, turned it on & let it bath overnight. Next morning - he found THIS:  It works , NUFF SAID.

ebay...ultrasonic jewlery cleaners. I'm working on winning a hot plate right now...then I'll be going for one of these. Liquid cultures I think will benifit even more from this...Agar..you should do up a batch of jars...some spores your regular way..some spores ultra soniced..some liq myc your regular way...some liq myc ultrasoniced..you get incredible results doing this as is so if there really is a difference we should see it there. Those 4 groups would make excellent pictures for TLG I'm sure too to show people with controls how it helps.
I'm sure I'll test it myself but I'm more of a liq culture person then spores..still havnt made a syringe yet..and I dont have the ultrasonic yet..I'll be trying it with and without with liquid myc once I do get one but hey I figured if you've got the time and the urge

Ebay. If you are patient? You can get a GOOD one with fairly large stainless steel tank, for cheap. Best get one that has a 2 quart or bigger container & stainless steel. Name brand on mine is BRANSON

I hear ya....BUT
My foaf does liquid culture with one of these.
Once so thick it will not stir any more. He pours it into one of these (sterilized).
Hit the blend button on pulse a few times & you have the HOTTEST - FASTEST INNOC in the world.
If you want even faster & less work. Just snip a whole stem off a big shroom grown invitro(bag or jar)so it is as CLEAN as possible, sink it into a peroxide bath for 15 / 20 minutes, remove & blend in sterile water.
Store for a week in jar.. to see if any contams show up & if not.... go for it.

That stainless steel container happens to be a WARING (Pro bartender). I got it at a 2nd hand resturant eqt & supply place for $20 (grin). Agar the frugal

Oh yeah I forgot about that..
Its hard to find that sort of thing though..ultrasonics can be had for a cheap price easier...well you should still knock up some jars with those spores at the same time as some prepared the same way but without being ultrasoniced. Something to compare to is what alot of the naysayers want to see.

You have my attention. I am doing liquid culture, (karo), from spores, and own a huge ultrasonic cleaner.
What, exactly, should I be doing?

All you do is run it through the ultrasonic which vibrates it apart. Not sure how long it will take...anywhere from 30 seconds to 24 hours I'd imagine. If it works like spore dispursal then you should have an extremly broken up liquid culture.
This could also benifit growth somehow...either running it 5 minutes a day to agitate or perhaps even constantly for constant agitation. I'm not sure of what if any this would do but it could do good and IMO is worth experimenting with.
