OK guys' I made 36 pf tek brf jars with the 1/2 inch of dry vermiculite on top. I was going to try to use a few fully colonized jars and just transfer some of the colonized mycelium to these jars as opposed to a spore syringe. I do have a glove box but as a practical matter I'll have to dig through the dry vermiculite to get the mycelium deep enough in the jar. I know I should have used wbs or rye but I was just wondering if there is any practical way to do it with these jars as prepared. The mycelium syringe tek sounds doeable but when putting a chunk of colonized mycelium in sterile water you end up with a lot of vermiculite and substrate floating around which is hard to push through a needle. Actually getting a chunk of colonized mycelium into the jars seems to be the way to go, if it's doeable without contaminating everything.

why don't you fruit the cakes and get a spore print?

take the spores and make a karo jar... liquid myc solution.

or, pick your best mushroom out of the fruiting and make an isolate...

I have many spore prints and prepared syringes, just wanted to try the mycelium transfer tek. I think however that it would work best with wbs or rye. Prepared brf jars seem too open to contamination even using a glove box. Biggest problem is the 1/2 inch layer of dry vermiculite on top.

I don't understand why you would want to transfer myc to karo when you could just inoculate the karo with spores, hassle free.

No I was talking about transferring the mycelium directly to the prepared brf PF tek style jars. I know this is done with wbs and rye just never heard of it being done with PF tek style jars.

The karo tek is when you cultivate a myc culture on liquid. Kinda like a hydro myc culture. You mix water with light corn syrup (I believe karo is a brand of it or something) and the myc feeds off of the sugars of the corn syrup and grows.

For your grain to grain transfer:
Wipe down a larger ziploc bag (gallon bag or so) with alcohol or bleach or something to sterilize it then empty your BRF cake into it and break it up the best that you can. Then take the spawn and mix it with your jars. Stamets, the master, says that you can mix spawn to substrate at a ration or 1:10 to 1:40, with 1:40 being a little on the side of not being successful, but 1:10 is great anyway. Take about a tablespoon of spawn and mix it per quart with your freshly sterilized substrate. Excercise the greatest sterility possible when doing this. Check out the FAQ. If that doesn't help then do a search "G2G" or "Grain to grain transfers" and follow those procedures.

Sterilize some karo water, and suck it up into a sterilized syringe.
Then, shoot the water into the jar w/ the cake that is colonized.
Shake it up real well, then hold the jar on its side and suck up the water again.
You may now use this myc water to inocc you jars.
(But, you should probably plan ahead better next time...)

Thanks Jimber510, sounds like a plan. My thinking is that the mycelium syringe will colonize faster than the spore syringes which were averaging anywhere from 4 to 6 weeks to completely colonize.

With a myc syringe, you will get faster colonization times because the spores dont have to germinate. Also, you can knock them up w/ more fluid (myc syringes arn't as valuable as spore ones)
