Can myc conquer wetspot?

Hello folks

I am suspicious to one of my WBS jars that it has tiny pieces of wetspot.
I was wondering if the myc is aggresive enough can it conquer and overrule the wetspot?
Or is wetspot = doomed and toss it?

depending on amount of the jar already colonized....a friend has had myc conquer i on wbs before, but the jar was like 95% colonized....and he said when he broke the wbs and smelled/examnied it there were no traces--fruits were tasty too he said.

Majority of the time its a gonner....

i would wait and see. sometimes over time the wet spot drys out and myc conquers. i waited close to a month b4 my whole jars where covered in myc and the wet spot dried out im guessing cause of the hight temp in my growth chamber.

Hello Gema,

That is good and hopefull news! Wetspot (I suspect) is only in the upper part of the jar and only on a few grains per spot.
The jar is about 40-50% colonized now with mycelium, It started fast I hope it will continue.

those don't look much like wetspot... but the sour smell is enough to get me and i doubt the myc. will win that battle.

toss it. i'd clean with bleach because i'm a clean freak when it comes to my grow.

Its been out the incubator almost a week now. I cleaned it with vinagre/h2o2.
It smells real bad/sour disgusting. I think it IS wetspot but slightly, some grains have that brown wetlook around them which the myc cant colonize it seems.

that sucks, try using tyvek to decrease moisture in your jars.

Hello Gema,

As far as I know wetspot is not created/triggered by too moist environment. It makes the grain look wet by itself. It a tough bacteria which can survive the PC if I read that correct.

All the other wbs jars (same batch) didnot have it so IMHO its not the moist. Also it was colonising very good, when the jar is too wet the myc wont grow that fast.

Also the water is at the bottom but the wetspot was at the top, maybe I was not sterile enough?

remember that just because it's fully colonized with the desired species doesn't mean you have a pure culture. i've had cultures that seemed fine and healthy until i opened them up and smelled them.

I do see what could be wetspot on a few grains...it looks like the grains are mushing out against the jar but its creamy yellow...that stuff. It could just be burst grains but the color is different and reminds me of contam. The sour smell is also a bad indication.

I wash my contamed jars by hand and PC at 15psi for 10 minutes.

Looks like wet spot to me, and the smell is a dead give away that something isn't right. The myc. probably wont win that battle. At the very least, I'd isolate those jars and watch them for a while. GL with the rest of them.

That's for sure wet spot in those pics. It will NOT survive PCing if you make sure you are at pressure for the propper amount of time the whole time. Too wet is no good but IME it is introduced with your inoculation whether it be agar wedge, liquid or spore water. If your using spore water maybe you didn't flame the needle well enough before doing those jars. Anyway this is a fairly old thread so your probably done with that batch.
BTW if you have a few kernels among a fully colonized jar you might be able to overcome the wetspot and that's if you just case the grain. If your going to spawn straw, your doomed with any sign of wetspot.

I had it when the jar was 50% finished, could have tried to remove it. It was in the top.

But a matter a fact it is TRUE that the UNGERMINATED (or how I should call it) wetspot is very resistant to PC.
One of the reasons to soak!

The ugly smell, im trying to surive the jar in a cold place wanna put it in my compostheap when it is spring..

(And wetspot makes infected myc look wet but too wet grain is not the cause, maybe it helps a little?) :wink:

Wetspot is a bacteria, not a mold spore or fungus. There is no need to "Germinate" bacteria by soaking. All bacteria is destroyed by PC'ing. Now if you have a large volume of grain in your PC and do not increase your sterilization time accordingly, you may not be reaching the proper sterilization requirements. Up your PC time and there isn't a single bit of wetspot that can survive. Truth is wetspot usually can be traced back to a contammed syringe or liquid culture. Grain to grain transfers never get it unless your donor grain spawn has it already.

I dont think it was the syringe. Have inoculated other jars which did not contam.

And:
"The most practical method for eliminating bacterial endospores involves soaking the grain at room temperature 12 - 24 hours prior to sterilization. Endospores, if viable, will germinate within that time frame and then be susceptible to standard sterilization procedures"

A good soak helps, ofcourse increasing PC time too. But 90 minutes is what I do.

Did not have any wetspot anymore though.
