inoculation tip
    #3537008 - 12/23/04 08:50 PM (11 years, 11 months ago)

my method for cooling my needle after flame sterilizing is to fill the cap of the needle with alcohol and rinse it out then fill again and after the needle is red hot i just put it back in the cap to cool off before i inoculate, is this a good idea at all or is there something bad about doing this? suggestions welcome

That's perfectly fine. It shouldn't hurt anything provided you give the alcohol time to evaporate a bit before continuing with the inoculation. It is however, uneccesary because surgical steel heats and cools extremely fast. You can heat the needle to glowing red hot and it will be cool enough to touch agaisnt your skin within only a few seconds after removing the heat source. Don't go trying this and burn yourself though. My point is that it cools very quickly. If you must cool it off manually I suggest wiping it with an alcohol wipe or sterile cotton swap that has been soaked in alcohol after you heat it. The way you suggest works fine as well though and I've heard of people using this method. Moreso for the sake of sterility than cooling, however.

thanks for the advice again mr. juice, i think the cap makes a nice place to put the needle in between jars. i thought about the alcohol maybe still being on the needle and killing the spores when i inoculate but so far ive done 15 like that with no problem.
