Have I screwed up?

I filled a syringe full of boiling water and let it cool. The syringe and needle where straight out of their, "Sterile untill opened" packet. 2 hrs later. I got a shot glass, cleaned it with disinfectant, rinsed it and rubber it with 70% Isopryl (sp) rubbing alcohol. I scraped off the spores with the needle into the shotglass, but I'm so smashed I forgot to flame-sterilze the needle. It was straight out of the pack though, and had been imersed in boiling water for a while. As soon as I relized that I flamed it. Then I continued to scrape the spores off the index paper then sucked back all the water into the syringe. I can't see any difference in it. I can't see any spores and the water is the same color, but I'd scraped off the whole print. There was actually tiny, tiny bits of the paper floating in there. Does any of this matter? You should do allright. You don?t have to see the spores in the water, since they are microscopicaly small. If you sucked the spore suspension back in the syringe, you will be ok. It should be no problem not flaming the needle before start, if you didn?t touch it with bare hands.

Thats the problem with prints on paper, paper fibres end up in the mix. But if the print paper was sterilized properl;y then you should be ok. One thing though, if the package said 'sterile', then the needle is good to go. Boiling it, flaming it, screwing around uneccessarilly with it is only inviting trouble! Overall, your method should work although I prefer to sterilize things in the pressure cooker and leave the chemicals out of it.

When I say 'straight' out of the packet, I mean straight after I sucked 'em full of boiling water and let them cool down on a towel for two hours. I hadn't touched the needle with my hands after it had been imersed in boiling water, and it had the guard on for those two hours.

when i do my syringes i dont put the needle on the syringe untill i innoculate... i just fill the syringe with boiling water, let cool... use my wire loop to scrape spores into the empty (and sterile) tall shot glass and squirt the water on top of the spores and suck it right back up... i do it as fast as i can... then i let the syringe sit in the fridge until im ready to innoculate the jars... then when i innoculate i open the needle from the package and put it on the syringe... and continue on with whatever tek im using... that way theres no chance for the needle to become unsterile...
