Makeing a flow hood
    #3482462 - 12/11/04 04:22 PM (19 years, 3 months ago)

OK I've been thinking about making one of these for a couple years now, am i right by saying to make one you need a filter on one side then a blower then another filter you work in the Flow of air. it seems like a simple concept.

Can some one give me a good like to a cheap construction of one of these.

Also what is the beast hepa filter you could use.

Flow hoods are no big deal. They can be easily made w/ prefilter usually charcoal, the blower as discribed in the link, then Hepa filter. it is all quite simple.

There is another option you might consider. check out this clear tote box set up. It provides sterile (reasonably sterile) positive static pressure inside as it forces the clean air out arm holes:

I'm reasonably sure I have a couple extra new Hepa filters out in the shop I could trade for prints of enoki or maitaki or?

What makes you think this? What kind of a flow hood is it? Did you test it in a way to see wheter it is or it isn't sterile?

Try opening clean agar plates in front of the hood for 10 seconds and see if they contaminate.

g2g is not a very reliable test, what if the spawn was contaminated to start with?

Did you spray filter before you installed? Did you silicone seal everything? Did you run it 15 - 20 minutes before you did g2g. Were bags or jars & tops CLEAN on the outside? One HARD sneeze can make a differance. Could have been any number of things.

Sterile culture technique can be tricky sometimes. I always shake my masters the days before transfers and judge the ones I will use by how healthy they recover. A good point Agar brought up was the important issue of letting that HEPA run 15-20min. prior to purge the flowhood and immediate area of contams. also there are only three things that can contam your efforts besides a contamed master, number one is you yourself, number two is your equipment, and number three believe it or not is your flowhood from either improper maintanence or design. I've seen some peeps build a flowhood with a 95% particulate arrestors I can't even call them a HEPA I also see peeps using 97.99%  which IMHO is a terrible waste of a flowhood as introducing a large amount of 3% contaminated airflow say over a still air glovebox just doesn't make alot of sense in my book. One very important point I'll reiterate as much as I can is that you DO NOT want a high airflow coming from your HEPA a gentle breeze is what you want creating as little turbulence as possible   Like agar said it could be a number of things and a flawless sterile culture technique isn't always as easy as you would think. looks like: power inlet into one side, power outlet to ventilator.
the more speed, the more power, the more volume getting pumped.

but thats just judged by how it looks, i never had one o these in my hand.
peace ohm :mushroom2:
