ok guys i started my jars using the z strain about two weeks ago, i didnt get them into the tnt incubator until the second week. out of four jars only one is really beginning to colonize. if the other ones dont come along should i do a grain to grain transfer with them? or should i start all over and make some new jars and then a transfer. there are no signs of contam on any of the jars. by the way i am using wbs. thanks.

i dont understand this transfer you are talking about. the only way i see it possible is if you have a glove box. they should be good and give them about 2 weeks before you start freaking out

It's an enclosed box with gloves attached that allow for sterile work.

it allows you to do all your work in a sterile environment. ie: inoculate. if you dont have it then dont transfer. actually dont transfer at all. i dont know where you got this idea but i've never heard of it. since they were not at the right temp then its possible not to see anything yet i would predict to see something in about 6 days.

i got the idea from one of the wbs tks on this sight. it shows colonized wbs being transfered into a jar on non colonized wbs.

A glovebox, or flowhood is going to be pretty much needed for any grain to grain transfer to work without contaminating. A glovebox is pretty much a really small cleanroom. It is most often made out of of a clear rubbermaid box with holes cut in it with a hot knife and gloves glued or taped really well to the holes. This way you are working in a small controlled area closed off from the outside world and all of it's contaminates. A flowhood is a workspace that is partly closed off with air being forced over everything, often times with a blower of some sort, that has gone through a HEPA filtration system so that you know that it is all clean. If you search the posts you will find hundreds of ways to build either, cost from almost nothing to hundreds of dollars.

While z strain is often times reported as very fast to colonized over others many factors should still be taken into effect. Often times the reports of times are done with karo-water which goes much faster over spore syringes due to the fact that the karo water has already gone through the germination processes.

yeah karo is the shit. it not only speeds up colonizing time it cuts down on the chance of contams.

i just tried the karo tek with some pan cyan the karo has alot of growth but the wbs has nothing its been about 7 days

yeah i meant the karo my bad. hmmmm it works like a charm for me but ill tell you exactly what i do. i stick the needle in suck up fluid and then shoot it back in the jar really fast this breaks it up real good. but to be honest i dont know shit about pan cyan. and the normal wait time to see mycelium forming is 5-7 days for cubes. so im guessing it to be a time thing.

yay its my first pan cyan grow and its its my first wbs grow probably not a good combo. but oh well the only thing i can think of is i didnt soak the wbs the tek didnt say to. i though u had to soak to activate the endospores

this is a little out of my league so dont take what i tell you to seriously. but im gonna guess you hit it on the head. i think its soak for 12 hours then simmer, never done wbs myself though. i think you can just soak for 24 hours also. once again i dont know but i think its so you can break open the millet so your spores can get there nutrients. if nobody tells you on this post i would suggest making a new post to get a more accurate answer.
