spores from dried caps...

i posted this question a couple of days ago, and i got some good ideas, and now ive got some kind of plan, which id like to get some feedback on at this point:

i have plenty of dried, closed caps (i figure closed cap has a MUCH smaller chance of contamination). i would like to (in a sterile setting), cut open a cap, remove some gill material with sterile tweezers and blade, break it up into dust, and soak this in sterile water for a new syringe or two. i know this may not be ideal, but i live in CA, and it doesnt seem that easy to get spores here. like i said, i have PLENTY of dried specimens to use for this. will this work?

someone also suggested i inject sterile water into a closed cap, but id think that the tek above would get more spores from the dried gills.

someone else suggested using agar to culture a dried gill, and pick a winner from that to use for spores. i like the sound of this, yet my patience would prefer a more direct approach.

ANYBODY with any experience using dried caps for spores please share your thoughts. does anyone think my plan at the top would give ok to good results? let me know, & ill love you forever.

Hey egghead ... when's that last time you grew anything from DRIED samples??? i think you just missed that part;).... closed caps have immature spores... peace

so am i screwed then? a few people have told me, in different ways, that i COULD use dried caps to get spores from. (i have closed & open, in abundence)

any suggestions here would be great...

ok ... here you go...

If your going to try your had at innoculation from these fruits your going to need 99% luck and the other 1% is tek..
basicly what you want to do is ok ... but i wouldn't suggest throwing in crumbled up gills... see if you can tap some spores off of the mature fruits.. if not then , as you said, your screwed... peace and good luck ....

Not necessarily.
Some(most?) strains begin to drop spores shorly before the cap opens.
So placing a gill on agar would still be a possible successful move.

yes i would think "closed caps" means immature fruits... a cap that is showing it's veil attached to the rim and the stem obviously starts sporulation because it's about to start doing it. it doesnt' necissarily mean that the cap is closed...i think it's just that you dont like me that your picking apart this poor souls post...

I have been successful both times I have tried making syringes from dried caps and I am about to do my third attempt with a new type. What I did was flame sterilize a needle tip and wipe with alcohol swab. Scrape the inner surface of the cap into a sterilized dish. This should produce a bit of dark powdery material. You don't need too much and less gill material is better. Add sterile water to the dish and mix with sterile needle. Draw in the water from the dish into your syringe and hope for the best. I usually let my syringe soak over night before inoculate, but it is likely to also work right away. Seems better to let soak 24 hours though. The important part is keeping the dish, water, syringe and needle sterile.
