...assuming that that the liquid culture was less than 2 weeks old, or do you need to use tissue, spores, or a solid substrate or somethin?

mycelium grows. Suck up some liquid culture and squirt it into a new jar of karo. I can't see why this wouldn't work.

I hear say of potency loss, but I can't understand it with taking some mycelium and giving a new place to grow. :confused:

Hrmm, can you think of a better method to grow mushrooms then than using a perpetuated supply of liquid culture and spawning it to bulk? You don't even need a PC!

Sure. You can take a sample from 1 karo jar and transfer it to another. It will only be a clone if the 1st karo jar was started with cloned material, though.

As I understand it, it has to do with age. Cells are only designed to replicate themselves so many times before, during the replication process, errors occour and you have genetic degredation.

Got any clue as to how many generations it takes before that happens?

Also, I have cheap Meijer brand honey, but it says "Pure Clover Honey US Grade A" and on the back there are no indications of additives or additional ingredients at all. Would this work for a liquid culture?

I don't know how long it takes for a fungal cell to replicate itself, but I heard on the radio or something that a cell divides fifty times before it dies.

The program went on to explain that there is research into extending that number indefinately. Aparently in the early part of genetic research someone thought they found the secret, and it pissed off every scientist who tried to copy his experiement because they couldn't get their cultures to survive. I guess the original experiment was completely bunk because it wasn't immortal cells, just regular reproduction, but no one figured that out for fifty years.

Surely that jumble of layspeak clears everything up :wink:

If it says it's pure honey, it shouldn't have anything else in it.

Why do you want to use honey, though? I was under the impression that it is less desirable than karo, because honey is colored and contains proteins and other substances that can further cloud the mixture, making it more difficult to discern whether you've got mycelium or contamination.

Karo is just a brand name of light corn syrup.

You can pick up a bottle of generic light corn syrup for less than 2$ at any store that carries baking supplies.

That was after 2 weeks. I've shaken it a few times, but it doesen't do anything. They just swirl around in there... lol.

I wasn't under the impression that it matters? I haven't used that jar yet, but when I do.. I will just use a syringe and sucking/spitting action to break the balls up before the final draw.

I tried making some karo just recently. I've been stirring it everyday and I see many tiny clusters forming all over. Nothing like that

Yeah. I have two karo jars of that strain. The 1st jar is pictured, the 2nd jar is quite a bit different.. It has many more thinner, smaller.. more wispy blobs. Swirling that one around does indeed break them up a bit.

I believe you should give at least 1.5 weeks before taking a sample. You want to make sure there is plenty left in there to re-colonize.

You can make them as big as you want, as long as they will fit into your PC, really... but there's no reason to go nuts and make a quart jar of karo or anything wacky, lol. It'd be better to make many smaller samples, so you don't lose all if one gets contaminated.
