Humidity outside of the jar would have no effect on humidity inside the jar because the jar should be air tight. If the substrate is mixed correctly there is no need to worry about humidity inside the jar during the colonization period. Though a temp of between 80 and 85 would speed colonization. Find the magick temp that works.

You two are right, ambient humidity has nothing to do with the process of invitro colonization. It is important for casing colonization but not substrate colonization in jars.

Psilocybe cubensis was happy to show that he doesn't know what he is talking about.

Probably beacause he speaks about bulk substrate with open type colonisation(compost trays...)
But sure, a much too low humidity will dry out any substrate on a longer turn.

If I am right jars alway's have the possibilty to breathe, so if rH inside the jar is higher than outside the jar will dry out in time. When colonising in 7 day's this isn't a problem. When doing it from spores (10 day's) and when temps drop it will take longer and the chance of drieng out rises. But at lower temps the proces takes longer (drying out) and it will turn out fine after all only if colonisation takes months but the chance of contamination would be large 2 than! So both are right! The possibility of drying out is present but will never take place because jars are colonised before this can happen.

Your jars are not going to dry out unless they are sitting in the middle of the 100 degree desert. Your cake will be ready to birth long before a temp of 80 degrees dries out your jar.

Do you keepn the tape on br/ver jars. After a week I noticed that jars colonize about 40% of the surface. Should I take the tape off for more gas exchange or will it dry out? Maybe just flip them over? Temps around 76-79 degrees stored in sterilite container

It depends on where you live. My friend has to be careful
with filter discs or the TOP of the jars tend to dry out.
She lives almost two miles up where it is very dry... the
relative humidity averages around 15%.

This is good but I like to add "you little dick" after each "grow" along with some of "Bluhoney's Mycoporn"
That should speed things up a bit, no doubt....lol

On a more serious note:
I innoculated pint jars on Sunday so by my count its been incubating for 4 days now and today I can positively identify spot colonization on all 6 jars. Question: I have 5 holes on each lid I innoc'd the shit of em with 1/2cc in the 4 normal holes and 1cc in the middle, I did not use any tape over the holes just aluminum foil. Should I keep the foil on the jars at this point or take em off for more air exchange? Im pretty much protected from contams with the dry verm correct?
