specific questions about the incubation process

if i use a lighter to sterilize, how long do u guys general sterilize it for (until it gets red hot?) and do u sterilize the entire length of the needle? i'm asking this because im about to re-do my first grow and my last 4 jars were all contams. i'm only going to inoculate 6/8 jars to try and isolate the contam issue.

also 3/8 of my jars contain about 60 cc water and the other 5 contain about 50 ccs. which ones do u guys think will colonize faster?

also if i have a hepa filter running in my room (it's dusty) is the air movement going to affect the colonization? i think i read somewhere that you're not supposed to have air flow when incubating.. maybe it meant just INSIDe the chamber?

one last question - does it matter if the jars are a bit warm to touch when innoculating? should they be at room temp?

i alcohol wipe the entire lengthe of the needle, and then heat it with a lighter at the tip allot, and only a little up towards the other end to make sure i burn the alcohol off. let the very tip get red hot, but dont keep it red hot or yull melt the plastic part of your syringe.

nobody can say what one will colonise faster.

do not inoculate your jars if its a little warm, if the glass is a little warm, the substrate may still be deadly hot for the spores.

i would run the hepa filter at all times, except when i am opening the incubator, or pmp type setup.

Air flow shouldn't really affect your jars that are incubating as they should only have small holes in the lid, and some people keep their foil on or use bandaids to cover the holes so I don't see how air flow could seriously affect them unless you put them in front of a fan with the holes pointing toward the oncoming air.

Psychoslut pretty much summed up the rest.

I'll just add onto the colonization speed part. Some spores could germinate faster than others and also some bits of myc will colonize much more rampantly (rhizos) than other fluffy myc. So it's all just kind of random which one will colonize faster as Psycho said.
