Potato Dextrose Experiment

Just wanted to make sure it worked, so I did a little experiment.

Hypothesis:

Homemade Potato Dextrose Solution synthesized from potato dextrose and water will hold nutritional content for growth of bio organisms.

Materials:

1 Kerr normal mouth quart jar
Mirro 8qrt Pressure Canner
Wash rag
2 coffee filters
400 grams of old potatoes
2 quarts tap water

Procedure:

1) Pilled 400gs of older potatoes and cut them in 3mm slices.
2) Boiled with 2quarts tap water for 50-70 minutes
3) Sifted through a rag hanging conically in a quart jar
4) Filled the quart jar half full
5) Placed a coffee filter underneath the lid and a 1/4inch hole.
6) Screwed on band loosely
7) Covered with aluminum foil
8) Pressure cooked at 15 p.s.i. For 30 minutes
9) Placed on top of a hand towel in a paper bag in a warm dark closet
10) Incubated in warm dark place
11) inoculated with a live unsterile tissue
12) Lightly shook jars every 12 hours

Data:

4 hours - No noticeable growth.
12 hours - Liquid?s opaqueness is increasing, white growth on bottum of jar
24 hours - Small blotches of white at the top of jar, liquid is almost opaque
36 hours - Noticable odor from jar, white growth around the top of the liquid, completely on the bottum
48 hours - Growth generally everywhere. Disposed of liquid for safety precautions.

man, i made some PDA mix that I didn't pc, as i was saving it until friday to do. this was just the other day but i checked it today and it was nasty. frothy white crap all over and sort of a sick sweet smell to it.

So wait, you're saying you tried to do a liquid culture with a PD solution, and it worked fine?

Of course why wouldnt it? PDA is just a liquid culture with agar added so you can take slices. Anything you could make agar with would work as liquid culture. Including water used when simmering grains..water you run through poo ect. Liquid culture is just nute+water sterilized and knocked up. Any nute cubies will grow on can go in the water.
