Strain degeneration?

I don't know the language of this science well enough to do a good job of asking this question. Sorry.

The teks say that you can use part of a colonized jar to make a mycelium culture and thereby inoculate other jars.

Can this be done repeatedly over generations without wearing out the strain? In other words, each batch provides the mycelium for the next.

I wouldnt go past 3 generations... if you can pic up the Paul Stamets book

the mushroom cultivator, it will give you much more indepth info on cloning,
spawning and strain degredation.

Yes it will cause degeneration according to stamets. A general rule with grain to grain transfers is to keep it within 3 generations. The example used in TMC (Great book to get by the way!) is starting with 1 jar from spores (g1), using that jar to inoculate 10 jars (g2), and then using each of those jars to inoculate 10 more jars (g3). Therefore you can create 1000 qts. of colonized grain using just 1 jar! Now, Incubating all those jars is where the real challenge is... :thumbup:

I'll put the book on my Christmas list. Just to keep the newbie question ball rolling;

Can I stall those 10 jars for long term use in the refrigerator?

What keeps strains fresh? Tissue cultures? Spore prints?

I know, it's in the book. Just don't have the book yet.

you can make several tissue cultures from the first generation and store them on
agar under distilled water or mineral oil, water is the best. you can freeze them
in a deep freezer and they have been known to remain viable for as long as 20
years... under mineral oil 5 years is the expected life span, simply remove,
drain and thaw the culture and then transfer it to agar plates and spawn

another method is to take spore prints, when doing generational work, vary the
substrate used at least every 2 generations. spore prints can remain viable for years as well
