Hello dear people,

I have been asking a lot, and I certainly will be the time ahead.

So I would like to have your advice once more on the subject of sterilizing all attributes.

In the teks one can read different stuff to desinfect/sterilize with.
Such as Lysol, Alcohol, H2O2.

But what is the best to desinfect:
- inoculation chamber
- fruiting chamber
- airstone & hoses/airlines (how is that called in english? :smile: )
- pressure cooking the scalpel every time might suck
- Casing containers
- Thermometer / hydrometer
- Syringe needle (A flame is dirty I read!?)
- My hands!
- Vermiculite / Geolite (I doubt that it always comes sterile, bags of geolite have small holes in them I saw at our local store)

How many of you do use labgloves and/or mouthfilter (lol dont know english name)

Ok a whole list maybe the answer is simple I would be happy to find out!

steralize the verm by popping it in the microwave (only when wet) boil the geolite.

Use anti-bacterial handsoap to wash all this. I use spray lysol to clean out my fruiting chamber/incubation chamber and then wipe it down real well.

For Vermiculite I turn my oven on 250 degrees and allow it to get up to temperature, put vermiculite on a baking pan and throw it in the oven for 15 minutes. Some people microwave theirs, but the verm gets so hot it melts plastic.

Syringe needle use an alcohol lamp, lighter, or isopropyl alcohol to wipe it.

For the thermometer use the misting lysol aerosol and then just wipe it up with a clean rag.

Airstone etc should be fine if you are using h2O2 in your water, but I imagine you could use lysol disinfectant spray on it as well.

When I'm working with cakes outside of the jar I use sterile hospital gloves, and also when I'm preparing my substrate or working with cakes about to be birthed I use actual medical face cloths. I also wear either a hat or a medical hair net most of the time when working in open air space.

Specially tips like hairnet and medical clothing, never thought of that!

Bleach isnt that the stuff they put in the toilet, like "chloride"?
Yes I want to put 1/10 H2O2 in the PMP.

Hey Aussie another question,

How much is your PC filled with geolite when boiling? Like 1/2 in 2 inches of water?
The PC I have (arrives Saturday) is 6,5 liter..... (13 pints) :P

So now I am going to write all info down creating mushroomfriends tek! :p Some things I read like 4 times but it is so much information that not everything stays....

Use my bathroom, see below
- fruiting chamber
Bleach in a 1:200 ratio to water (yes thats 200) and alcohol in a spray bottle
- airstone & hoses/airlines (how is that called in english? )
I don't sterilize it, you just pour h202 in the water..as long as nothigns growing the H202 will kill it all. You could fill a spray bottle with alcohol (I do, for the fruiting chamber and casing containers, utensils, whatever) and spray it with that. Would evap quickly
- pressure cooking the scalpel every time might suck
You only need to PC them for 15 minutes. Throw them in an empty jar to keep them out of the water. You can also flame sterilize scalpels and such tools and/or wipe down with alcohol, I use only alcohol for spoons and stuff to break up substrate and spoon out casing
- Casing containers
Alcohol in a spray bottle. Once I've used them once, at the end of the casing, I dump the casing left (after 3 harvests usually), wash it out kind of well with just warm water. Let it soak in warm soapy water for 24/7 (sometimes more or less, tin pans are harder to clean then plastic) and scrub well then goes through the dishwasher. Then just sits and gets the alcohol spray before use
- Thermometer / hydrometer
Don't worry about it
- Syringe needle (A flame is dirty I read!?)
Flames are not dirty. You would want to get an alcohol lamp (doesnt burn iso alcohol, you have to get denatured alcohol from a hardware store, the lamps will be there too) for best results as it won't leave a black residue. I use a regular lighter though and wipe the outside (and black off) with a cotton ball soaked in alcohol
- My hands!
Wash hands with anti bacterial soap, put gloves on, wash hands with it again.

How many of you do use labgloves and/or mouthfilter (lol dont know english name)

The english name would be surgical masks or paint masks. I use latex gloves (trich likes to hide under your finger nails) and a paint mask when working with my stuff always. I empty out the bathroom of all carpeting and towels..throw in 2 HEPA filters on high for a few hours..spray Oust! (or lysol) in the air, wipe down surfaces with Tilex (says it kills 99.9% of bacteria and mold). Then (having had a shower 2 hours sooner before cleaning the bathroom) I go in wearing only my boxers, gloves and the paint mask. Theres nothing cloth for spores to hide on, same for bacteria. Enter with all my jars and such and do grain to grain transfers, agar, knock up jars, casing, liquid cultures, printing/syring making, everything. I turn Both HEPAs off once I've entered and sprayed oust once again to prevent blowing spores around (HEPA's are only so efficient).

The thing is with stuff like Geolite, though it has holes in it, its non nutrious. Contams don't like it and so probably won't be there and wont germinate unless they have food anyway. Plus myc fights off contams naturally. Sterility is important but you don't need to clean EVERYTHING, like a therm/hygrometer that just goes in your chamber. or Geolite or perlite or wands. Just pour h202 in any water you use (I use it for dunks, spray bottles, all my humidifiers, everything) for your chamber. Good luck~!

PMP box yes, incubation no. The substrate should be sheilded by the Tyvek. If using the PF Tek you might want to though since foil and the dry verm barrier (no tape!) is your only barrier for contams. Thats enough but you should be a little more sterile in the incubation tub. Once again though its just plastic, no nutes, not easy for spores to stick to, not such a problem. Once again the spray bottle comes in handy for both of these. No problem!

Yes I use PF tek with verm layer on top of substrate in the jars.
In addition I want to use alu foil and if possible the lids over them while colonizing.

It should go..substrate in jars..verm layer up to top..lid with 4 holes in it..ring..tin foil. Remove tin foil to knock up and replace once done.

Lol that makes sense guys... I should put a ring between the lid and the alufoil?

Cant I just skip the foil and use bandaid on the holes instead, after my jar is pregnant? :laugh:
Must not put the lids too tight cause cake must be able to breath?

You can put the lids tight..foil goes over the ring because you want to be able to remove it easy. Thats the point of the tin foil and why you shouldnt cover the holes. Bandaids should work because they should allow your wounds to breathe but tinfoil is cheaper and easier to put around it. You definatly shouldnt need both..

You still need the ring with the band aid. The lids are two part..the lid (the flat part) and the ring (just holds the flat part on). You want tin foil or band aids over all that.

I thought a lid was ring+lid from on piece. The lock of the jar..
Hmmz nasty sometimes this international thinhy with languages... :crazy:

But I thought only covering the holes would do, at leats according to some different teks?
