The spawn is based on small woodchips that can be bought in a pet-store. It is used as animal bedding but can also be used to smoke fish and meat. These chips are made from beech.

these are put in the pressue cooker..
(we put a metal lid on top to prevent the bags from blocking the steam valve)

EXCELLENT stuff, UNA!!
The only question I have is.... Don?t you think 2 hours(not sure of PSI) is a little overkill?
My mother is a retired Microbacteriologist and SWEARS 45 min @15 psi will sterilize ANYthing.
Of course those bags are pretty thick, 2 hours is probably NOT such a bad Idea--it damn sure can?t hurt!

Two hours is certainly not overkill. I think it is the minimum time that is required for bags this big.
45 minutes is ok for liquids (maybe even a little bit too long) but for solids such as grain and supplemented woodcips longer sterilisation is required. Depending on the size of the container we sterilise between 1 and 2 hours.
No offense to your mother but 45 minutes will not do the trick

Yeah, it?s definitely different for woodchips than anything a bacteriology lab will usually deal with.
I think I remember reading in GGMM something about a recommendation for THREE hours for a certain supplementation of woodchips.
Those pics are phenomenal, just one question. . .

How do you get the moisture content of the chips before puttig them into the bags? Do you just wait until they stop dripping? How long is this on average?

This year I didn?t sterilize the woodchips (they were(looked) the same as Una?s)), I only soaked them in warm water for 3 hours, brought to boil, boiled 15 minutes and strained.
NO suplementation, just pure wood chips.
Once cool, innoculated with rye grain spawn.
No contamination problems.

Una, Myself nor my mother are offended:) However 45 min @15 psi will sterilize ANY thing(that we can think of)- I do understand that it would not suffice with your bags of substrate, not because of the substrate it?s self but rather the way that the amount of substrate is being sterilized.
Case and point... if you were to put the same amount of substrate in an autoclave and it was spread out to say an inch or two thick, it would be perfectly sterile in 45 min @ 15 psi.
I have been using rye-grain for about a year now in 1/2 pint jars that are used to colonize other substrates-- I preassure cook them for 45 min @15 psi and have not had any contam problems(while still in the jars) to date.
Anywho... This is a VERY nice post!! Very well documented! If it is not already, you should incorporate this into your site.

a friend has noticed a prob he had with yellow and black mold hitting jars. BUt he thinks it might have been some woodchips he missed that were in the birdseed, anyone else have this prob, either that or airborne or on a scalpel. THis has hit a quite a few jars from the ass end of a bag of penningtons. ANy ideas? anyone have contam from old lids? I soaked the jars in bleach water even.

ok...so my thread is turning into a "how long should you sterilise something before it's sterile"...

In think(and i do also know about microbiology and plant tissue culture) the required time depends on:

a) if it are liquids or solids

b) the amount of substrate in a container

Liquids require only brief sterilisation, solids require longer (beacuse liquids conduct heat much better, and don't have small cavities)

The more substrate is in a container, the longer it will take for the heat to penetrate to the core..

I believe that exposure to 121oC for 45 minutes will indeed sterilise anything.....but before the core of the substrate has reached that temperature may take quite some time.

I think you are both right...but you both mean a different thing....

VERY well said, Una. 45 min@ 15 psi WILL sterilize anything---BUT it all depends on density and mass---to achieve temps at the core of whatever is being sterilized times NEED to be adjusted.

Hi,
Don't you think adding oat is really necessary? I think one can get contams if does not work in really sterile environment. You shouldn't put your culture outdoor until spawn is fully colonized also... When you use pure woodchips, you don't have to keep it home and wait till all is colonized...

No but it speeds up colonisation dramatically. It also produces thicker and more luxurious mycelium. Instead of oatmeal you can also use bran.

True, but since we have a laminar flow cabinet (really sterile environment) this is no problem.

You shouldn't put your culture outdoor until spawn is fully colonized also...

No you shouldn't...Of course we let the spawn fully colonise before using it outdoors. Indeed if the woodchips are not supplemented putting them outdoors before 100% colonisation should pose less problems..

The spawn was used to spawn more small wood chips. These small chips were soaked for 24 hours. Also bigger pieces of wood (10 x 2 cm) were thrown in (these were soaked for some days).
I estimate that the bed is 25 cm deep.
